| Tillandsia is one kind of plants which don’t need to grow in the soil, and are able to growth in the air, can be managed easily and purify air condition. It has been favored by people more and more as ornamental, and its market prospects are very broad. But Tillandsias’roots are degenerated and have adhesion or fixed effect only, do not have the function of absorbing water and nutrition, rely only on blade to absorb water and nutrients in the air, so they grow very slowly. Both points of vegetative propagation and seed breeding, Tillandsias’propagation speed are extremely low, which greatly hindered its commercial production. Tillandsia was selected as the material for this study that explored the condition of tissue culture technology for accelerating its growth by configuring different culture medium and improved its growth by interval water spray. And the study also explored the diurnal variation analysis of the three kinds of air pineapple on CO2 absorption, determination of Tillandsia stricta storage and germination rate of seeds. Main results are as follows:Tillandsia stricta seeds in MS medium, sugar-free MS culture medium and single AGAR medium can grow seedlings, but on the MS culture medium seedlings grow faster significantly than in sugar-free on the MS culture medium and AGAR medium (p< 0.05). Tillandsia stricta seedlings cultured in MS medium with auxin (NAA and IBA) significantly grow faster than in MS medium without auxin and adding cytokinin (6-BA). According to the leaf number growth, the effects of different concentrations and types of auxin to promote seedlings rapid propagation is in the order:0.4 ppm NAA> 0.1 ppm NAA and 0.2 ppm IBA> 0.1 ppm IBA. Adding cytokinin in the MS medium has no boost effects for seedlings growth than single MS culture.With Tillandsia stricta leaves as explants transformation, using alcohol to disinfect 30s and then using 0.1% HgCl to disinfect 45s and the disinfecting effect is best with pollution rate is 0% and the survival rate is 100%. In MS medium respectively adding 0.2 ppm,0.4 ppm and 0.6 ppm completely matching combination of 6 - BA and NAA, can induce Tillandsia stricta leaves produce callus, and the rate in the range of 20-67%.Relatively. The 0.2 ppm 6-BA+0.8 ppm NAA combination has the best effect, and has the highest rate. Preliminary results show that, using the Tillandsia stricta leaves as explant transformation is feasible.Tillandsia spraying interval test showed that Tillandsia in both cases of short spraying interval and long spraying interval can grow, but very slowly, and under the condition of suitable spraying interval unit its increment is the largest. Tillandsia stricta seeds’ storage using freeze dry method, founding that seed germination rate in two months is slightly lower than just picking the seeds. |
