Photinia fraseri 'red robin' is the hybrid variety of Rosaceae Photinia.Considering it with the characteristic of strong ecological adaptability and admirableappreciation, it is used widely as important landscape plants.The research, using thestem of Photinia fraseri 'red robin' as experimental material, based on the tissueculture technology, the induction culture, proliferation culture and rooting culturewere studied, aiming to established and optimize the system of micro- propagationtechnology of Photinia fraseri 'Red Robin'. Which could make the theoreticalfoundation for application and industrialization of Photinia fraseri.The major results showed as flowing:(1)The optimum sterilizing condition is that explant was disinfected with75%ethanol for 15 seconds, then treated with 0.15% HgC12 for 8 minutes. Choosing thebuds in the third to sixth following the apical bud as explants, the budding rate is high,browning rate is relatively low. The sampling time were early time of March, Apriland September.(2)Using MS medim as the the basic, setting 27 hormone formula with 6-BA,NAA,KT and IAA, compared with other formula, the formula 6-BA and NAA hasobvious effect. Besides, the result also showed that hormone concentration in a certainrange could accelerate the growth, however excess hormone would restrain thegrowth.The optimum medium was MS+6-BA1.0 mg/L+NAA1.0 mg/L. the inductionrate was 95%.(3)Using MS medim as the the basic, setting 32 hormone formula with 6-BA, NAA,ZT and TDZ, The hormone TDZ and ZT both had the function for theproliferation of Photinia fraseri 'red robin', the proliferation coefficient would be 3when the medium added TDZ, and ZT was 1.7. In the same time, excess hormonewould restrain the growth. Obviously, the effect of TDZ on proliferation was betterthan ZT. Combined TDZ and ZT in the proliferation culture, significantly shorteningthe time of proliferation and improving the proliferation rate and proliferationcoefficient, having a obvious effect on proliferation, the combined use of TDZ and ZTcould shorten the time of subculture. The optimal proliferation medium was MS+TDZ0.05 mg/L+ZT 0.2 mg/L, and the proliferation rate reached 100%, the multiplicationcoefficient was 7.1.(4) In the rooting culture, adding NAA or IBA in the 1/2 MS medium had theeffect on rooting, compared with IBA, the effect of NAA was better, the growthcondition of root was better, the rooting rate could be 95%, which solved the problemof rooting of Photinia fraseri. Darkness culture 5 day, then turned to light trainingwhich contained the 2500 lux, 16 hours every day, could shorten the time rooting.Therefore, the suitable method was to take root dark training 5 day at the first, thenturned to light training, the medium was 1/2 MS + NAA 1.0 mg/L, the rooting ratewas 96%, and the average number of root was 18.(5) Setting different ratio of substrates, exploring the influence on the matrixsurvival rate, found that the survival rate of the mixed matrix which contained perlite,vermiculite and peat was higher than single matrix. Appropriate matrixwas pearlite:vermiculite: peat = 3:2:1 (V: V: V), the transplanting survival rate could be 93%.
