This research includes two parts.The taxonomy of Rhytismataceae on Angiosperm from Jianfengling National Forest Park was mainly studied using the traditional taxonomic method in the first part.After the sorting and inspection of the samples from Jianfengling National Forest Park and comparing them with specimen from the Forest Fungi Dried Reference Collection of Anhui Agricultural University(AAUF),13 species from 5 genera of Rhytismataceae parasiticing on Angiosperm were identified,including Coccomyces delta,C.dentatus,C.mucronatus,C.occultus,Hypoderma rubi,Lophodermium agathidis,L.camelliicola,L.conigenum,L.heterocladum sp.nov.,L.illiciicola,L.pinastri,Marthamyces chinensis sp.nov.,Terriera minor,of which Lophodermium heterocladum and Marthamyces chinensis were new species.They were preliminarily established by internal microexamination and external examination of frozen section,detail description of morphological features,internal structure characteristics and so on in both English and Chinese were given,and the point line composite image of the ascoma,ascus,ascus spore and paraphysis were given.The authors also investigated and studied in geographic distribution,ecological habitat of these species.In the two part study,combining the bioinformatics methods and molecular biology,based on ITS rDNA for the preliminary study,the phylogenetic trees were constructed to explore the intergenera,interspecies and intraspecies relationships of the Rhytismatales members from all over the country.DNA was extracted by CTAB method,and the correlation sequences was amplified by PCR and sequenced.Leotia lubrica as outgroup,phylogenetic trees were constructed respectively,combination with morphological classification method,the species classification status was determined.The results showed that the same species usually preferred to cluster together,but in some conditions belong to two different genera or species are also likely to gather together.This indicated that identification of fungus just relying on a sample kind of method was not credible,and a variety of biological technology was needed.If on the basis of morphological classification,combined their individual development,cultivate character,polygene analysis and so on,its phylogenetic relationships could be determined more accurately.As the advantage of accurate and fast,the molecular biology technology has played an important role in classified study,so we should improve the DNA barcoding technologyand optimize DNA database of fungus as soon as possible.For all this,the traditional morphology method is still the main reasonable basis in fungi classification.
