Pepper is one of the main vegetables in Shandong province. In recent years, with the maket demand for pepper increases and the pepper acreage expanding rapidly, pepper virus diseases have become a limitted factor in the pepper production. Most of viruses showed a mixed infection, pepper virus disease has become the main factor that cause the low production and poor quality, which restricts the development of the pepper planting industry.In the survey, based on molecular biology technology, pepper samples collected from 10 growing regions in Shandong Province were detected for the presence of 15 species of viruse pathogen. The occurrence and distribution of pepper viruses, the predominant viruses and mixed infection incidence in Shandong Province were evaluated preliminarily In addition,Shandong isloate PeVYV was cloned, sequenced and phylogenetic analyzed.The main conclusions are as follows:1.Using the 15 specific primer pairs to detect the 253 pepper samples which were collected from pepper producing areas of Shandong Province. 14 viral pathogens were detected. The viral pathogens were PMMoV, CMV, TMGMV, BBWV, BWYV, TMV, ToMV,TYLCV, PVY, MABYV, CABYV, PeVYV, AMV and ChiVMV. No sample positive to CaCV.BWYV, MABYV, CABYV and PeVYV infection were first reported in pepper plants in China.2.The virus infection rate was 96.0% for the total 253 pepper samples. Among the viral pathogens, PMMo V and CMV have the highest infection rate, were 61.6% and 60.0%respectively, which were the most important viral pathogens in Shandong Province. TMGMV,BBWV, BWYV and TMV were very common pathogens, the infection rate were 42.3%,34.3%, 24.9% and 24.5% respectively. BWYV is the newly reported virus pathogen.TMGMV become one of the main viruses infecting pepper in Shandong. The detection rate of ToMV, TYLCV, PVY, MABYV, CABYV, PeVYV, AMV and ChiVMV were 0.6%, 7.5%,6.7%, 3.5%, 2.3%, 1.9% and 0.3%.3.Among 243 positive pepper samples, only 34 samples were infected by single virus.209 samples were mix-infected with more than two viruses, the mix-infected rate was 86.0%.30.4% samples were infected with three viruses. The detection rate of the two viruses and the four virus mixed infection were 27.9% and 27.5%.4.The full genome of PeVYV was acquired by overlap PCR, The obtained nucleotide sequence of PeVYV in this study was consistent above 90.0% with the PeVYV reference sequences in GenBank.
