Pathogen Identification And Detection Of Pepper Virus Disease In Xinjiang

Abstract/Summary:
For a long time,chili virus disease has seriously affected the production and quality of pepper,which is one of the important reasons for the production reduction of pepper production.Due to the growing area of pepper planting and the frequent exchange of germplasm resources across the country,the species and quantity of pepper toxins are constantly changing.To clarify the occurrence and distribution of pepper virus diseases in Xinjiang,this study used metagenomic sequencing technology combined with molecular biological detection technology,452 pepper samples collected from 7 regions in Xinjiang were identified and detected for 20 pepper viruses.Using the reported seed-borne virus species and the primers that have identified the Xinjiang pepper virus species in this study,RT-PCR detection was carried out on the pepper seeds(seedlings)of 10 varieties(lines)on the market,the following results were obtained:1.The Illumina Hiseq sequencing platform was used for viral metagenomic sequencing.A total of 5pepper viruses were identified in the pepper samples,and the sequencing results were verified by RT-PCR.Among them,4 viruses have been reported to infect peppers in Xinjiang,namely Cucumber mosaic virus(CMV),Bell pepper endornavirus(BPEV),Pepper mild mottle virus(PMMo V)and Pepper cryptic virus 2(PCV2).It was the first time that Beet western yellows virus(BWYV)was found to infect Xinjiang peppers.2.Molecular identification of 20 viruses was carried out on 452 pepper samples collected from 7regions in Xinjiang,and a total of 9 viruses were identified,namely Broad bean wilt virus 2(BBWV2),BPEV,BWYV,CMV,PMMo V,Pepper cryptic virus 1(PCV1),PCV2,Potato virus Y(PVY),Tomato mosaic virus(ToMV),and successfully Clone into PCV1 whole genome sequence.For the first time,PCV1 and To MV were found to infect Xinjiang pepper.3.The detection rates of BBWV2,BPEV,BWYV,CMV,PMMoV,PCV1,PCV2,PVY and To MV in452 pepper samples were 9.29%,40.93%,7.08%,37.17%,34.51%,2.88%,34.29%,2.65%,0.88%.Among the detected viruses,BPEV had the highest detection rate of 40.93%,followed by CMV with 37.17% and ToMV with the lowest rate of 0.88%.Among the detected viruses,BPEV had the highest detection rate of40.93%,followed by CMV with 37.17% and To MV with the lowest rate of 0.88%.A total of 349 pepper samples were infected with viral diseases,accounting for 77.21% of the total samples,including single infection and co-infection.Among them,108 were single infection,accounting for 23.89% of the total samples,and 241 were compound infection,accounting for 53.32% of the total samples.It can be seen that the Xinjiang pepper virus is dominated by compound infection.There are 52 compound infection combinations,2,3,4,5,and 6 toxin compound infection combinations are 14,17,15,5,and 1,respectively,and 3 viruses are compounded.Most infestation combinations.The compound infection mainly consisted of five viruses,CMV,BPEV,PMMo V,PCV2 and BBWV2.Among the 52 combined infection combinations,the infection rate of BBWV2+PVY was the highest,which was 16.67%;the infection rate of CMV+BPEV was the second,which was 16%;and the infection rate of BPEV+BWYV+PCV2 was the lowest,which was only 0.66%.4.Detect 11 kinds of viruses including PMMoV,CMV,TMV,ToMV,ToMMV,CGMMV,BBWV2,BPEV,PCV1,PVCV2,BWYV2 on the seedlings of 10 pepper varieties(lines),and found 10 pepper varieties(lines)seeds All of them can carry the virus;the seeds of 10 pepper varieties(lines)all carry BPEV;there is a phenomenon of virus compound infection in the pepper seeds.
