In this study,through the vermiculite and nutrient solution culture experiment,we studied the effects on growth and physiochemical indexes of wheat?Triticum aestivum Linn.?after inoculated with Glomus mosseae under Pb and Cd stress,which include morphological changes,fresh weight and dry weight,Chlorophyll content,photosynthetic rate,heavy metal content of roots and leaves,autioxidant enzyme activities and production rate of O2?-,content of methylglyoxal and GSH,glyoxalase I activity.In addition,this study was also briefly analyzed the transcriptome of wheat under Pb stress.In order to explore the detoxification effects and mechanism of wheat after inoculated with Glomus mosseae under Pb and Cd stress,the results are as follows: 1 Effects of Glomus mosseae Inoculation on Detoxification Capacity of Wheat under Pb StressLow concentration of Pb could promote the growth of wheat,while high concentration of Pb had adverse effects on the wheat growth.When inoculated with Glomus mosseae under Pb stress,Pb content in root was increased and in the leaves reduced,fresh weight,dry weight,chlorophyll content and photosynthetic rate were all increased,the morphology was much better than equivalent stress concentration uninoculated wheat.These showed that inoculated with Glomus mosseae could promote the growth of wheat under Pb stress.Compared with the normal wheat,content of MDA,H2O2 and production rate of O2?-in leaves were significantly increased,in the meantime,the activity of SOD,POD,APX and CAT were significantly enhanced.While after inoculated with Glomus mosseae under Pb stress,content of MDA,H2O2 and production rate of O2?-and the activity of SOD,POD,APX and CAT had varying degrees decline,compared with equal stress concentrations of uninoculated.These illustrated that inoculated with Glomus mosseae could effectively reduce the oxidative damage caused by Pb stress.Compared with the normal wheat,the content of methylglyoxal and GSH were increased significantly under Pb stress;glyoxalase I activity was increased under low Pb concentration stress,but decreased under high Pb concentration stress.After inoculated with Glomus mosseae under Pb stress glyoxalase I activity was increased compared with equal stress concentrations of uninoculated and content of methylglyoxal and GSH were both decrease.According to the results of wheat genome transcriptome analysis,there were 1031 genes expression quantities increased and 951 genes expression quantities decreased under Pb stress;after inoculatied with Glomus mosseae,there were 700 genes expression quantities increased and 1429 genes expression quantities decreased under Pb stress.Then,through the comprehensive analysis of the transcriptome clustering relations,genes relations,the main effect genes and selected noteworthy enrichment of GO and pathway,finding that the differences of the growth and metabolic between wheat inoculatied and uninoculatied with Glomus mosseae under Pb stress were caused by some genes related to the physiological and biochemical processes,mainly including plant hormone regulation,amino acid metabolism and transport,cellular redox balance,carbohydrate metabolism,carbohydrate metabolism,glutathione metabolism,etc.2 Effects of Glomus mosseae Inoculation on Detoxification Capacity of Wheat under Cd StressLow concentration of Cd could promote the growth of wheat,but high concentration of Cd brings adverse effects to the wheat growth.However when inoculated with Glomus mosseae under Cd stress,Cd content in leaves was reduced and in root increased,fresh weight,dry weight,chlorophyll content and photosynthetic rate were all increased,the morphology was much better than equivalent stress concentration uninoculated wheat.These showed that inoculated with Glomus mosseae could promote the growth of wheat under Cd stress.Compared with the normal wheat,content of MDA,H2O2 and production rate of O2?-in leaves was significantly increased,at the same time,the activity of SOD,POD,APX and CAT were significantly enhanced.While after inoculated with Glomus mosseae under Cd stress,content of MDA,H2O2 and production rate of O2?-and the activity of SOD,POD,APX and CAT had varying degrees decline,compared with equal stress concentrations of uninoculated.These illustrated that inoculated with Glomus mosseae could effectively reduce the oxidative damage caused by Cd stress.Compared with the normal wheat,the content of methylglyoxal and GSH were increased significantly under Cd stress;glyoxalase I activity changed little under low Cd concentration stress,but decreased under high Cd concentration stress.After inoculated with Glomus mosseae under Cd stress glyoxalase I activity was increased compared with equal stress concentrations of uninoculated and content of methylglyoxal and GSH were both decreased.
