Grape(Vitis L.)as one of the most important fruits of economic crops in the world,has an important position in the production of fruit trees in the world.Grape is also important fruit tree in our country,our country is the great powers of viticulture,according to the international organization for grape and wine(OIV)statistics,in2014,the Chinese grape planting area of 799000 hectares,since 2000,China's grape acreage than total global planting area has risen from 3.9% to 10.6%.In terms of the world,eighty percent of the grapes used for processing,our country is given priority to with table.Resveratrol(Res),as a polyphenols in plant.The study of Res dating back to the middle of last century,now has found that Res there are at least 12 families,31 genera and 72 species of plants,many of which can be used as human food,such as grapes,giant knotweed,cocoa,beans.Res is considered to a phytoalexin be produced by fungal infection,mechanical injury,the ultraviolet irradiation.Later because the resveratrol has the discovery of significant physiological functions in plant and animal body so it gets people's more attention.Grape is a major source get Res for human,studies have shown that high Res content grape is usually the wild grapes,has poor edible quality,and the current production on a large number of cultivated Eurasian species and hybrids in Europe and America under the condition of natural Res content is low,the Res content in its peel and seeds is usually lower than 2 ug/g FW,it cannot satisfy the requirement of Res.This experiment by using HPLC method was developed for the determination of96 is given priority to with wild grapes of resveratrol in the skin of grape germplasm resources and,on the basis of the combination of SSR technology content high Res genome-wide association analysis of related genes,explore the RS alleles associated with Res,to explore,to protect the wild grape excellent genetic source,lay the theoretical basis of grape Res biosynthesis,promote functional stability and sustainable development for the grape industry.The main results were as follows:1.Found some higher levels of Res content in grape berry skin,the wild grapes: ‘V.adenoclada Shuangxi 03',‘V.adenoclada Zhijiangshui',‘Tangwei seedling',‘V.adenoclada Shuangxi 01',grape varieties: ‘Zijixin',‘Yalishanda',‘Miskat Plevenski',‘Zaotianmeiguixiang',‘Fenghuang 51 hao',‘Xiangfei'.2.Found some higher levels of Res content in leaves,the wild grapes: ‘Gaoshan No.2',‘V.ficifolia Fengjugou 03',‘V.davidii Wuhan',‘V.betulifolia Songxian',‘Vitis pseudoreticulata 1057',grape varieties: ‘Zifeng',‘Champion',‘Irsay Oliver',‘Zhengguoliuhao',‘87-1',‘Zhengzhouzaoyu'.3.Determination of wild grape germplasm resources in about 60% of the strains of the skin in the Res content is higher than that of leaf blade,grape cultivated in about 38% of the strains(kind of)blade Res content is higher than that of the skin4.For wild grapes,meteorological conditions have obvious differences in two years,under the condition of different years of the same change range of grape peel Res content between two small,annual performance between content stability;Different vintages from the same strain Res content in leaf appear difference5.Culture content of resveratrol in grape skin more sensitive to the change of the weather,the concentration of two in table grapes results show a significant difference between the two,in late July 2013,picking of the grape peel Res content is higher than the same period in 2014,and in early August 2013 harvest grape peel Res content is lower than the same period in 2014.Content of resveratrol in grape leaf varieties also show that the sensitivity to climate change,mature leaf collection on July 15,the grape varieties leaves Res content is significantly higher than in 2014,2013,causes of this phenomenon may be early in July 2013,zhengzhou region last half wet weather.6.With the content of grape Res data with SSR markers we on the behavior of grape Res content has carried on the correlation analysis,and the content of the grape peel Res associated tag has Sh74,Sh53,Sh71,including Sh74 in 2013 and 2014 in the two years of analysis related to the peel Res phenotypic explain rate were higher than 5%;Associated with content of grape leaves Res tag has Sh74,Sh28,Sh58,Sh68,Sh74,including Sh28 in 2013 and 2014 in the two years of analysis related to the blade Res of phenotypic explain rate were higher than 5%.Combined with two years of Res content,primers Sh74L(Forward primer: 5 '-ACTCTACTTCCTATGGACGAA-3',Reverse primer: 5 '-GTTGTTGTTGTTGTTTGCAG-3')is a significant control the skin and the QTL of the resveratrol content in the leaves,as well as between the year of the existence of stable control of Res in QTL primers Sh28(Forward primer: : 5 '-TCTCAATAAACCAACTCT-3',Reverse primer: 5 '-ACGTGGCTCTTATAATAC-3').
