In recent years, people found that androgens and anabolic steroids can significantly increase the output of animal muscle, so it is widely used in animal production. This kind of medicine has the bigger side effect, in vivo metabolism time is longer, it has an important influence on the health of the human body and the development of minor, so it has caused people pay more attention to analysis of the residue. So it is great significance to detect and analyze the residues in animal foods.In order to establish a rapid and efficient method for the detection of androgen and anabolic steroids in livestock and poultry meat. Also to study the experiment conditions of pretreatment method, the condition of the high performance liquid chromatography and mass spectrometry were explored and analyzed. The method has been selected QuEChERS method as the pretreatment methods, detection method for high performance liquid chromatography tandem mass spectrometry. In this study, the extraction solvent, the conditions of purification, mobile phase composition, the conditions of mass spectrometry are the main factors. The test has chosen beef, mutton, pork and chicken liver as samples in Gansu Province. Study the linear relationship, recovery rate and repeatability of this method, and through the practical application in meat safety testing verifies the reliability and practicability of the method.. The main results as follows:(1) Establishment of detection method of LC-MS/MS:Collecting standard mixed solution’s signals via tandem mass spectrum MRM mode; using positive ion scanning mode, ZORBAX Eclipse Plus Cig chromatographic column was determined to be used for separation, the flow rate was 0.25 mL·min-1, and the column temperature was 30℃, the recovery of spiked samples was 51.06% to 91.34%, the relative standard deviation was 0.64% to 3.62%, the detection limit was 1 ng·kg-1; the limit of quantitation was 100 ng·kg-1 to 300 ng·kg-1.(2) Optimization of pretreatment methods:Traditional pre-treatment methods usually need to be enzymatic hydrolysis and to maintain more than 12 h in the 37℃. Used QuEChERS method which was based on SPE. Compared with SPE, this pretreatment process was simplified, greatly shorten the time, the whole sample treatment process takes only 20 to 30 min and provides the theoretical basis for the rapid determination of androgens and anabolic steroids residues in animal foods.(3) Sample detection:In 97 samples,4 samples were detected testosterone residues, the total positive rate was 4.124%; 1 sample was detected trenbolone acetate residues, the total positive rate was 1.031%. Show that the method can be used for the detection of animal meat in the hormone residues. Its sensitivity, accuracy and precision are in line with the requirements, and pretreatment was simple.The experimental results show that this method is suitable for the detection of androgens and anabolic steroids in meat of livestock and poultry, which can avoid the false positive results and has good practicability in the actual production.
