Intracerebroventricular injection, a new drug delivery technology, has great practice value in clinical trials. It can not only be used for treating many brain diseases, but also widely used to study the brain ventricle formation and morphogenesis, and describe the mutant brain morphogenesis. Objective: To establish the rat model of bacterial meningitis and compare the therapeutic activity of penicillin following 3 consecutive days' intracerebroventricularly administration or intravenous injection at different dose level. Method: A total of 16 male SD rat, approximately 250-300 g of bodyweight, were divided into 4 experimental groups. All the animals were anesthetized and inoculated with Actinobacillus pleuropneumoniae(APP) to establish the rat bacterial meningitis model. Then the animals in drug treatment groups received 3 consecutive days penicillin treatment following intracerebroventricularly administration or intravenous injection at different dose level. The clinical observation, histopatholog, bacterial culture and Polymerase chain reaction(PCR) were utilized as the important index for pharmacodynamic evaluation. Results: According to the typical symptoms and the result of PCR test result, we succeed to establish the rat bacterial meningitis model. Then the sick animals were subjected to various treatments following established dose regimen. In the following drug treatment phase, compare with the model control group, the drug treatment lessened the clinical symptoms of the disease. The curative effect of animals received intracerebroventricular administrations of penicillin at 0.01 mpk was better than animals received IV administrations of penicillin at 10 mpk, and was roughly the same as the animals received IV administrations of penicillin at 62.5mpk. Conclusion: Rat bacterial meningitis model was established in current study. Intracerebroventricular injection of penicillin is an ideal method in treating bacterial meningitis, the dosage is lowness and efficacy is good.
