At present, Peanuts are the fourth largest planting crops in Jilin province. In recent years, Due to the global climate change, Drought occurred frequently, Results in the decrease of crop yield greatly. Peanut is drought, poor crops, But also the first crop of drought disaster avoidance. This test for 260 peanut germplasm material, combined with morphological characters investigation and SSR marker analysis, build the primary core germplasm; And evaluate primary core germplasm for drought resistance in seedling stage, Put forward based on Li- 6400 portable photosynthesis device without damage of appraisal indicators. The main research results are as follows:1. Based on the selected 40 core SSR primers, Building the 260 peanut germplasm DNA fingerprinting and genetic diversity analysis, The experimental results show that:(1) Selected 40 on core SSR primers were detected 139 allele, Each pair of SSR primers amplification of allele number 2-7, on average is 3.56; The range of variation of Polymorphic information content(PIC) is 0.1225-0.750, on average is 0.5123.(2) Using the software which named NTSYS2.1 to make a UPGMA Cluster analysis,The results show that,the 260 materials were divided into 4 group which stand by A,B,C,D. The variate nuclear primer could completely distinguished the whole materials.(3) Using the software named MEGA4,Rebuilding the phylogenetic tree for 260 materials based on the basis of NEI genetic distance(GD).Construct the evolutionary tree by means of NJ,ME and UPGMA,The result shows that 260 kinds of materails could be divided into 4 germplasms in genetic evolute relationship.and its divide result is as same as the UPGMA Cluster result.2. Utilizing 260 parts of peanut as research materials, and use its 14 quantitative charaters to construct a peanut modal database, The experimental results show that:(1)The Euclidean distance was superior to Mahalanobis distance, The cluster by the way of single linkage method was better than the furthest neighbor linkage, the middle distance method,the centre-of-gravity method, the group average method, the variable class average method, the flexible method, the sum of squared residuals method. The preferred sampling method was superior to the divergence from sampling method and the random sampling method.(2)With 50% sampling ratio, Take advantage of euclidean, then combine the preferred sampling method with the single linkage to make a cluster analysis, and elect 128 samples for building core collection,(3)At the same time, use the SSR molecular marker technology to analyze these germplasm’s genetic diversity, The result shows that 139 allelomorph were detected out in total in the 40 pairs of SSR primer.each pair of SSR primer amplified 2-7 allelomorph, on average is 3.49. The range of variation of polymorphic information content is 0.7499-0.0854.on average is 0.5065, Represent 80% of the total germplasm genetic information. With this clarification, the core collection which built by means of quantitative character and molecular marker was able to stand both original germplasm’s character and its genotype.3. In a simulated drought identification conditions, By the way of analyze the drought tolerance of 128 core collection, The experimental results show that: The number of drought hardiness sample we screened out between extremely strong and strong was 1, drought hardiness strong was 25, between strong and moderate was 53, the moderate was 36, between the moderate and the feelble was 13. these records established a basis for the drought hardiness identification of peanut seeding stage.4. This test make use of the Li-6400 portable photosynthesis system and SDPA- 502 plus chlorophyll meter for the first time to carry on the non-damage system identification during the seeding stage of peanut, The experimental results show that:(1)Finally analyze the measured data, and it turned out that Photosynthetic, Excitation energy capture efficiently of PSII reactive center which exoteric to the light, Photochemical quenching coefficient, Photochemical quantum yield at the aspects that stress treatment has relatively obvious amplitude of variation, they are also fit for evaluate the drought hardiness of peanut. Can be used as seedling stage without damage identification index.(2) Through the SDPA-502 plus chlorophyll meter measured SDPA values are not suitable for peanut seedling drought resistance identification.
