Campylobacter jejuni and Salmonella both are zoonotic pathogens in nature that mainly cause gastroenteritis, diarrhea and other diseases in animals and humans. Poorly handled or improperly cooked poultry meat, raw milk, pets, and untreated water are thought to be sources of infection. Bird is the main storage where people and animals infect Campylobacter jejuni and Salmonella. More and more drug-resistance bacteria appear caused by Abuse and unreasonable application of antibiotics, and drug-resistance Campylobacter jejuni and Salmonella make a big threat to human health.In this paper, Medias was screened to embedded lactic acid bacteria into probiotics by vacuum freeze- drying. The best embedding material was identified as: soluble starch 100 g / L, skimmed milk powder 30 g / L and corn starch 20 g / L. Probiotics viable count was up to 2.33 × 1010 cfu / pb after freeze-drying, probiotics have several characteristics either, such as a high viable organism release rate, moisture resistance, have a long-term preservation at-20 ? and 4 ? and about 30 days at room temperature(25 ~ 30 ?).The results of established Campylobacter jejuni and Salmonella Real-time PCR standard curve:(1). Linear relationship curve expression between recombinant plasmid copy number(Copynumber) and linear expression versus threshold cycle(Ct) of Campylobacter jejuni is Ct =-3.52 x + 47.92, correlation coefficient is R2 = 0.996.(2) Linear relationship curve expression between recombinant plasmid copy number(Copynumber) and linear expression versus threshold cycle(Ct) of Salmonella is Ct =-3.43 x + 43.2, correlation coefficient is R2 = 0.994. These two standard curves both have a high similar coefficient and small error.In this study, Experimental group was received the basal feed with 1.0%(w/w) probiotic, and the basal feed of control group with nothing. The growth performance, immune function and intestinal mucosa of chicken were detected during the experiment. The results showed that: The average weight of experimental group was Increased by 9% compared to control group(P ? 0.05). The fabricius index and thymus index of experimental group Increased by 10.44%?36.56%(P?0.05) separately at 25 days. fabricius index, spleen index and thymus index Increased by 40.12%, 19.30% and 36.57% separately at 35 days(P?0.05). The intestinal wall and the intestinal mucosa of small intestine in experimental group were thicker than that of control group, intestinal villus in experimental group were more longer and thicker, compared to control group. These results suggest that probiotics could increase body weight and immunity, and promote the growth of gut in chicken.In order to study the function of probiotics in prevention and treatment of Intestinal diseases, chickens were divided into 4 groups, each group receive different treatments. The results showed that: after attact with Campylobacter jejuni ZC8 in 11 days, the average weight of prevention group were 30.83 g, 78.57 g and 14.39 g higher than that of the control group, virus attack and treatment group in 29 days. Treatment group was 64.18 g higher than poison attack group; discharge amount of Campylobacter jejuni in prevention group showed a downward trend compare with poison attack group. After receiving probiotics treatment, discharge amount of Campylobacter jejuni in the treatment group drop down compare with poison attack group. After attack with Salmonella typhi CMCC50115 in 19 days, the average weight of the prevention group had reduced by 9.0g(P> 0.05) compare with the control group in 29 days, improved by 176.2g(P <0.01) compare with the poison attack group. Discharge amount of Salmonella in prevention group showed a downward trend compare with poison attack group, while the poison attack group, by contrast, the discharge amount of Salmonella reached 1.64 × 105 cfu / g(P < 0.01). After receiving probiotics treatment, discharge amount of Salmonella in the treatment group drop down compare with poison attack group. These results illustrate that Probiotics has a obvious function on prevention and treatment of Campylobacter jejuni and Salmonella typhi disease.
