Manglietia grandis,evergreen broad-leaved tree, belongs to the family Magnoliaceae, which is endemic to China.In the IUCN Red List of Threatened Species, the gandis is classified as “Very Endangered” plant.Manglietia grandis has beautiful outline,big and bright flower, good quality wood which are with highly ornamental value and economic value. Therefore, a great understanding of breeding technique can be beneficial to the deployment and dissemination of Manglietia grandis.This paper carried out the seeds' morphology and germination characteristics,container nursery techniques, grafting propagation techniques, tissue culture techniques of Manglietia grandis, and the results are as followings:1?Manglietia grandis' seeds with 9.77 mm length, 8.71 mm width, 4.42 mm thickness and 143.61 g grain weight. There is little difference between seeds..2 ? The germination rate of Manglietia grandis' seeds was 62.2%, the germination potential was 33.3%, and the germination index was 2.06. The seed dormancy is proved.3?The seeds that are storaged at room temperature can normally germinate and the germination rate is 60%. The seeds can be sown at the process of storage.4?Effective methods of germination : The Manglietia grandis' seeds were soaked by1000 mg.L-1 GA3 for 24 h, about 7 days later, they begin to germinate and finish in 27 days,which the germination rate reaches 71.1%. When the temperature is 25 ?, it is conducive to seed germination whose rate reaches 62%, the effect of light time on seeds germination was not significant..5?The better substrate for Manglietia grandis' seeding are peat: perlite = 3: 2.It is good for its seeding growth.6?The grafting propagation techniques of Manglietia grandis are initially established.It is with highly survival rate that reached 80% when Manglietia grandis was gfafted on2-year-old Michelia champaca in February. And 1 year later, it could reach the height of87.9 cm, and the basal diameter could reach 28.11 mm. The grafted Manglietia grandis can bloom in April in the next year. It grew faster than the seeding.8?This paper choose Manglietia grandis' seeds as explants and establish the Tissue Culture techniques. Suitable explants are with length of 0.5 ~ 1.0 cm radicle, they shoud be washed by 1% Bromogeramine 2 min and 0.1% Mercuric chloride 20 s, they grew better in the solid medium. The suitable induction medium was: MS + 6-BA 0.5 mg.L-1 + NAA0.05 mg.L-1, and the inductive rate is 83.3%. The more appropriate proliferation medium is:Improved MS + 6-BA 0.8 mg.L-1 + NAA 0.08 mg.L-1, and the proliferation coefficient is3.12.
