Momordicins compounds are a series of tetracyclic triterpeoids, which are isolated from the leaves of Momordica charantia and momordicins usually have biological activity to a variety of agricultural pests including Momordicin ?, Momordicin ?,momordicoside L and so on. Many studies showed that these momordicins compounds have neigative effects on the feeding, spawning and growth development of pests.But the effects of momordicins compounds on O. furnacalis larvae plasma proteins expression have not been reported.By measuring the effects of n-butyl alcohol extracts of M. charantia leaves and Momordicine ?on O. furnacalis larvae weight gain and energetic contents, the study detemined that the inbibition of growth and development for pests is the mode of action of momordicins. This paper aims to explore the inhibition mechanism of momordicins compounds on growth development of O. furnacalis and provide a scientific basis for the controlling of O. furnacalis and the developing of a new insecticides. The inhibitory effects of Momordicin?and Momordicin?on phenoloxidase activity of O. furnacalis in vivo were determined. And Two-dimendional electrophoresis were used to measure the effects on the expression of plasma protein.1. Effects of Momordica n-butanol extract on energy metabolism of O. furnacalisThe effects of n-butyl alcohol extracts of M. charantia leaves on energy substance contents were investigated by adding compounds to the artificial diet provided to O.furnacalis under laboratory conditions. The inhibition rate of weight gain of Momordican-butanol extract increased with the increasing of the concentration of the compounds.Treating larvae with alcohol extracts of M. charantia leaves for 3 days, the hemolymph total protein levels of larvae were decreased, the hemolymph total sugar were increased but the content of lipid of the treatments and the control were not significant.2. Effects of momordicin ?on energetic contents of O. furnacalisThe effects of momordicin ? on larvae energy substance contents were measured by feeding larvae with treated artificial diet and the results showed that momordicin ? had a significant inhibitory effect on the increase of larvae weight gain. Momordicin ? treatment can significantly reduce the hemolymph total protein levels of larvae, but momordicin ?treatment hardly influenced the hemolymph total sugar contents.3. Effects of momordicin?and momordicin? on phenoloxidase activity of O. furnacalis in vivoThe phenoloxidase of O. furnacalis was purified by precipitation with ammonium sulfate deposition. And the purification multiple was 2.50 and the recovery rate of enzyme was92.61%. Microplate reader was applied in the experiment by using L-DOPA as substrate.First, the effects of several common organic solvents on phenoloxidase activity in vitro were measured, including acetone, methanol, ethanol and dimethyl sulfoxide. The results showed that in the tested organic solvents, dimethyl sulfoxide had a minimal effect to A phenoloxidase activity when dimethyl sulfoxide content is 0 to 7.5%, phenoloxidase relative activity in vitro of O. furnacalis in more than 97%. Further, we determined the inhibition of momordicin ?, momordicin ?, momordicoside L and other compounds on phenoloxidase activity in vitro with 5% dimethyl sulfoxide as solvent. The results show that momordicin ? and momordicin ? had an obvious inhibition for the isolated phenoloxidase activity, which inhibiting concentrations(IC50) were 180.45 ?g/m L and 1126?g/m L respectively, and the inhibitory effects of momordicin ? was stronger than the inhibition of momordicin ?.4. Effects of momordicin?and momordicin? on expression plasma protein of O.furnacalisThe effects of momordicin ? and momordicin ? on O. furnacalis larvae plasma protein were measured by using two-dimensional electrophoresis. The results showed that the momordicin ? and momordicin ? also had obvious inhibitory effect on plasma protein expression of O. furnacalis. The O. furnacalis plasma protein of protein spots treated by momordicin ? and momordicin ? were significantly less than that of the control group, which reduced 226 and 331 respectively. Five spots were significantly different between the momordicin ? treatment group and the control group which were all increased. The 4th spot was identified as Keratin. Four spots were significantly different between the momordicin ? treatment group and the control group which were all increased. The 7th spot was identified as E-race odorant receptor 4.
