The poultry are frequently contaminated with Salmonella and can serve as vehicles in transmitting salmonellosis to humans through the food chain and it is also the most important foodborne disease in the worldwide. Therefore, it's very important to do some research about the distribution and spread of Salmonella in yellow-feather broiler slaughter processing and sales part(Broiler farms- Every main part of slaughterhouse-Market).In this study, a total of 800 samples were collected from every main part of yellow-feather broiler integrator producer chain(Broiler farms- Every main part of slaughterhouse- Market) in Guangdong province, and 108 Salmonella strains were isolated. The results showed that the contamination rate of Salmonella was different in every part of production chain. The precooling water samples from slaughterhouse was not isolated any Salmonella strains, the contamination rate of was low in the carcass viscerate part and downstream retail stores, 2%(2/100) and 7%(7/100) respectively. The high contamination rate was in chicken market, which was 66.67%(40/60). The contamination rate in meat market was especially higher than sales part of production chain, which indicated that every part of production chain had food safety risk issues. In this study, we found the different contamination rate in different time within the same part and also different contamination rate in the same time in each part of production chain. After deep investigation, we found this slaughter plants using recycling water, which was easily contaminated by Salmonella after using for a long time. The Salmonella contamination rates suddenly rose sometimes indicated that the microbial control program was instable in the slaughterhouse and we should further strengthen internal management of the slaughter plant.The isolated strains of Salmonella belonged to 11 serotypes, most of them were S. Indiana(45.37%) and S. corvallis(24.07%). Different part of production chain had different serotype distribution. S. Indiana was the predominant strain in broiler farm, but there was no superiority serotype in slaughterhouse. S. Oslo(100.00%) was the predominant strain in the cold fresh chicken sales market. S. corvallis(65.00%) was the predominant strain in the meat market around the slaughterhouse.The Salmonella isolates were tested for antimicrobial susceptibility against 15 antimicrobial agents by K-B method. The results showed that these isolated strains were highly resistance to Ampicillin(46.30%), gentamicin(39.81%), streptomycin(65.74%), nalidixic acid(52.78%), ciprofloxacin(42.59%), ofloxacin(38.89%), tetracycline(74.07%), chloramphenicol(46.30%), sulfagan(79.63%), trimethoprim(48.15%) and cotrimoxazole(48.15%), while these strains were sensitive to cefotaxime(98.15%), cefepime(99.07%) and ceftazidime(99.07%). The multi-drug resistance and phenotype was seriously and common, 74.07%(80) of the isolates were resistant to three or three more antimicrobial agents, and 108 Salmonella strains produced a total of 32 different resistant phenotypes.The chromosomes DNA of isolates was digested with restriction endonuclease Xba I and analyzed using PFGE. The PFGE patterns show that the same serovar Salmonella strains was distributed in the same big gene cluster, and the similarity was from 50% to 100%, which indicate that there was a high prevalent correlation of Salmonella in yellow-feather broiler slaughterhouse and retail markets. There were 17 PFGE patterns from the yellow-feather broiler slaughter processing and sales part at an 85% similarity threshold. Same Salmonella strain was isolated in each part of slaughter production indicated that it was existing the phenomenon of Salmonella spread along the production chain; some clonal strains(this strain had the same PFGE spectral pattern and 100% similarity) isolated from the same part indicated that the existence of cross contamination phenomenon in the yellow-feather broiler slaughter processing and sales part; The same sources of Salmonella could have different PFGE pattern, which indicated that the genotype of Salmonella was complex and more sources of pollution in the yellow-feather broiler slaughter processing and sales part.
