Isolation And Identification Of Corynebacterium Pseudotuberculosis From White Goat In South Shaanxi,and Development Of An Inactivated Vaccine With Aluminum Hydroxide Adjuvant

Ovine pseudotuberculosis also called caseous lymphadenitis,which is chronic zoonoses caused by Corynebacterium pseudotuberculosis.The disease is frequent in sheep and goats.It can cause sick thin, low productivity and stillbirth, great harm to the sheep industry.Shiquan county is located in the city of Shaanxi Province Health West, North Qinling Mountains, South pillow of Bashan, very suitable for Shannan white goat breeding, has been one of the city health Baishan goat farming county, 64119 slaughter Shaannan white goat only in 2014. In recent years, Shiquan County sheep farmers raised the Shaanxi reflect some white goat sheep appear in the upper part of a forelimb "Nongbao", various methods of treatment effect is poor, persist in sheep, causing large economic losses. In this paper, the incidence of the disease was investigated, diagnosis and prevention and treatment of the test, the following results were obtained.(1)Clinical survey results showed that the feeding capacity of a random sample of more than 50 in captivity and raised the big 10 captive, ram, ewe and lamb "Nongbao" average incidence rates were 46.4%, 38.1% and 1.5%, the backyard of ram, ewe and lamb "Nongbao" average incidence rates were 69.6%, 59.1% and 2.9%.(2)Isolation and culture of 30 strains of suspected pseudo tuberculosis in 2 samples from the diseased sheep were randomly taken. Application of bacterial 16 S rRNA universal primers for PCR amplification and sequence analysis showed that the isolated bacteria and Corynebacterium pseudotuberculosis 16 S rRNA homology 99.5%, identified isolates were Corynebacterium pseudotuberculosis, the disease was diagnosed as sheep pseudo tuberculosis.(3) The results of drug sensitivity test showed that the bacteria to ciprofloxacin,ceftriaxone, erythromycin, kanamycin and cefotaxime, gentamicin, tobramycin, doxycycline,chloramphenicol and other antibiotics are highly sensitive; moderately sensitive to tetracycline, rifampicin and streptomycin etc. Clinical trial results showed that cefotaximesodium, rifampicin and after subcutaneous injections, good treatment effect on sheep pseudo tuberculosis.(4) The inactivated aluminum gel vaccine was developed using the isolated Corynebacterium pseudotuberculosis. In safety and sterility test under normal circumstances,the results of small-scale field trials showed that in a year’s time, 30 sheep vaccinated only 1sheep sheep suffering from pseudo tuberculosis in 3.3%, the incidence rate, the protection rate was 96.7%, and the injection of sterile saline control group of 30 sheep in 23 sheep sheep suffering from pseudo tuberculosis, the incidence rate was 76.7%. Description of the development of the vaccine against goat pseudo tuberculosis has a good prevention effect.
