The Ralstonia solanacearum avirulent strain which used to prevent bacterial wilt of tomato is a feasible biological means. However, when discussed the culture condition of the Ralstonia solanacearum avirulent strains, we only think about single factor, not neglect the interaction under multiple factors. In order to explore the effect of interaction and the optimum culture conditions of the Ralstonia solanacearum avirulent strain against ralstonia solanacearum virulent strain, we used the suspected bacterial wilt of tomato as raw material to identify it and to explore the optimum and ideal culture conditions of the growth and the Ralstonia solanacearum avirulent strain against Ralstonia solanacearum virulent strain by the multifactor. The results as follows:1、The suspected bacteria which cultured on TZC medium showed that the edge of colony was white, the middle of colony was red and the colony has a lot of mucus, so the whole of colony showed pink and has fortissimo mobility;The suspected bacteria which smeared microscopy looked like rhabditiform; The tomato plants which inoculated the suspected bacteria showed a series of change that first the top leaves has wilted, then the bottom leaves and finally the middle leaves in the daytime and returned to normal at night after the three day. After a few days no longer resumed. Therefore the suspected bacteria was the Ralstonia solanacearum virulent strain.2、When we used the three factors together,the culture condition of the Ralstonia solanacearum avirulent strain which reached the maximum concentration was that the T was 35℃, the PH was 6,the culture time was 48h,.3、When we only focued on the single factor,the optimum culture condition of the ralstonia solanacearum avirulent strain with the maximum inhibition zone for ralstonia solanacearum virulent strain was that the T was 35℃,the PH was 6,the culture time was 48h; However, because of the interaction effects by three factors, the optimum culture condition of the ralstonia solanacearum avirulent strain with the maximum inhibition zone for ralstonia solanacearum virulent strain was that the T was 35℃,the PH was 7,the culture time was 36h, the ideal culture condition was that the T was 35℃,the PH was6-7,the culture time was 36h-48h.4、In this experiment, we also observed that the growth of the ralstonia solanacearum avirulent strain was positive correlation to the antibacterial ability. So there was a certain relationship between the concentration and the antibacterial ability of ralstonia solanacearum avirulent strain. |
