According to records, There were 14 Hemerocallis species worldwide, and about 11 species distributed in China. Hemerocallis varieties played a very important role in the landscape around the world.therefore, investigation,exploitation,utilization,innovation of Hemerocallis, had been an important research area in the horticultural circles.To promote the green application and germplasm resources innovation of Hemerocallis, in this study,the phenotypic traits of 75 varieties of Hemerocallis were researched; 13 Hemerocallis varieties were screened out to determine their ploidy, and 2 varieties of they were selected for karyotype analysis; The digital fingerprint and genetic relationship of 13 varieties of Hemerocallis were researched by SRAP markers. The main findings were as follows:1. The phenophase and morphological traits of Hemerocallis were observed and recorded,we could find it that Hemerocallis began to sprout in early-March, began to leaf in mid-March, went into the yellow period in mid-December. More than 80% of Hemerocallis went into the early flowering in June, flowering in June and July,late flowering in July; There was no secondary flowering in most Hemerocallis. Hemerocallis petal were mainly yellow, red and flesh, Secondly purple. There was no-eye in more than half of Hemerocallis. The pollen of Hemerocallis were mainly yellow and orange, the anther of Hemerocallis were mainly purple, white and brown,the Hemerocallis pistil were mainly yellow and white.There were mostly "star" flower-shape and "cup" pattern flower-model in flowers.We found it that there were some variation in Hemerocallis phenotypic traits which could indicate extensive variability of Hemerocallis germplasm through measuring and calculating.2. Analysis on phenotypic trait of Hemerocallis by correlation analysis, principal component analysis and cluster analysis, the results showed that there was positive correlation between Hemerocallis 11 main characters;all information expressed in 35 original traits variables could be approximately replaced by top ten main components.3. Determining chromosome ploidy of 13 Hemerocallis varieties by conventional root tip tabletting,we could find it that "Xiang Fei','Little Bee','Rose Convolution','Pardon Me','W Mini Pearl','Butter Rolls','Mary's Tone','Light Orange and Yellow','Frans Hals','Gemstar' were diploid,'Middle Red 1','V','Baltimore Oriole' ware tetraploid.4. The karytotypes of'W Mini Pearll' and 'Light Orange and Yellow' were analyzed,The results showed that the chromosome amounts of 'W Mini Pearl' were 2n=22, the karyotype formula was 2n=2x=22=20m+2sm, karyotype asymmetry index was 57.41%, belonging to 1B type of Stabbings karyotypic symmetry,relative length of chromosome was 2n=22=2L+8M2+8M1+4S; the chromosome amounts of 'Light Orange and Yellow' were 2n=22, the karyotype formula was 2n=2x=22=18m+4sm, karyotype asymmetry index was 58.28%, belonging to 1B type of Stabbings karyotypic symmetry, relative length of chromosome was 2n=22=2L+8M2+10Ml+2S.5. Six pairs of primer combinations selected from 4x4 positive and negative primer combinations were used to amplify 81 clear bands which include 73 polymorphic bands, 90.1% of the total bands. It indicated that the polymorphic in Hemerocallis varieties were rich,which could be used to reveal genetic relationship between 13 Hemerocallis varieties in the molecular level.6. Establishing digital fingerprint of 13 Hemerocallis varieties based on the primer pair me3-em2,that is,'Xiang Fei'= 000001001011001,'Little Bee'= 111011000100010,'Rose Convolution'= 011000000100110,'Pardon Me'= 100010000101100,'W Mini Pearl'= 011001111100010,'Middle Red 1'= 100001010101100,'V'= 100001000101100,'Butter Rolls'= 110111110101110,'Mary's Tone'= 011110001101110,'Baltimore Oriole'= 110101010101110,'Light Orange and Yellow'= 111101101101110,'Frans Hals'= 110111100100110,'Gemstar1= 110111010101110.7.13 Hemerocallis varieties were clustered based on SRAP markers, we found it that Hemerocallis varieties were divided into five categories when similarity coefficient was 0.68,the genetic relationship between Hemerocallis varieties of each category was similar. The result was consistent with appearance and ploidy of Hemerocallis.The above research could lay the theoretical foundation of species distinction, genetic relationship exploration,germplasm innovation of Hemerocallis,at the same time,it could provide the theoretical reference for relationship between the different genetic notation, gene mapping of Hemerocallis trait,molecular marker-assisted breeding and so on.
