Two previous reported species:Eimeria gruis, E. reichenowi and a wait taxon: Eimeria sp. were described in 76 of 164 Grus nigricollis fecal samples from Zhaotong Daoshanbao Black Neck Crane National Nature Reserve. E. gruis, which has the most prevalence of infection, was found in 35.9%(59/164); E. reichenowi-was behind E. gruis which found in 31.7%(52/164); the detecction of Eimeria sp. was lowest, which was found in 28.7%(47/164).According to 18S rRNA gene sequence of three coccidian in Grus nigricollis:(1) The identity of 18S rRNA gene sequence of E. gruis was 99.1?100% in this study. The homology of E. gruis was most close to E. gruis from Grus japonensis in GenBank (99.5-100% identity). (2) The identity of 18S rRNA gene sequence in the study of E. reichenowi was 99.9?100%. The homology of E. reichenowi was most close to E. reichenowi from Grus japonensis in GenBank (100% identity). (3) The identity of 18S rRNA gene in the study of Eimeria sp. was 100%. The homology of Eimeria sp. was most close to E. reichenowi from Grus japonensis in GenBank (99.2% identity). The homology between E. gruis with E. reichenowi in this study was 98.1?98.7%; between E. gruis with Eimeria sp. was 98.3-98.9%; between E. reichenowi with Eimeria sp. was 97.7?97.8%.According to Cytochrome c oxidase I gene sequence of three coccidian in Grus nigricollis:(1) The identity of coxl nucleotide sequence of E. gruis was 99.1?100% in this study. The homology of E. gruis was most close to Isospora amphiboluri from Pogona vitticeps in GenBank (81.8% identity). (2) The intraspecific identity of coxl nucleotide sequence of E. reichenowi was 99.8?100%. The homology of E. reichenowi was most close to Eimeria flavescens from domestic rabbits in GenBank (79.9% identity). (3) The intraspecific identity of coxl nucleotide sequence of Eimeria sp. was 99.6%. The homology of E. gruis was most close to Isospora amphiboluri from Pogona vitticeps in GenBank (81.9% identity).The homology between E. gruis with E. reichenowi in this study was 91.5?91.9%; between Eimeria sp. with E. gruis was 93.9?94.6%; between Eimeria sp. with E. reichenowi was 90.8?91.3%.In the present study, the species and molecular markers of coccidian in Grus nigricollis have been provided for the first time. The phylogenetic analysis of 18S rRNA gene and the coxl gene partial sequence revealed that the 3 species of Eimeria in Grus nigricollis were placed into a single branch in the tree.
