This research unifies indoor examination through the pot experiment, on the basis of grafting flue-cured tobacco root system reconstruction, explores grafting tobacco growth, tobacco alkali accumulation, drought resistance and anti-low temperature change, physiological mechanism. The main result is as follows:(1)Makes the scion by flue-cured tobacco variety K326, use solanum torvum as root stock conducts the grafting experiment, the grafting combination survival percentage reaches 91.2%, survival is good, the grafting efficiency is high. At the same time under the condition of not ridging, K326/solanum torvum grafting leaves contain no nicotine accumulation of tobacco plant, and leaf shape, leaf dry weight have no significant difference compares K326 natural tobacco plants and K326/K326 grafting, the growth potential is good.(2)In dealing with the ridging induced adventitious roots. K326 natural tobacco plants and K326/K326 grafting, under different processing conditions were not significant difference between the indicators. And K326/solanum torvum grafting compared with the other two kinds of tobacco leaves of tobacco plant. Under no ridging conditions, the leaves of tobacco plant leaf length, leaf width, the amount of dry matter and chlorophyll content difference was not significant, but plant height, stem circumference, dry matter allocation rate, nitrate reductase activity, nicotine content exist significant difference; After moulding processing, the difference among the three kinds of tobacco within high, stem encircles reduces gradually, but the adventitious root and main root spot's dry material distribution factor and root system vigor's difference gradually increases, nitrate reductase activity and nicotine content is still exist significant difference, the K326/solanum torvum grafting of adventitious roots of tobacco plant dry weight and root vigor significantly greater than the other two kinds of tobacco plant, but the nitrate reductase activity and nicotine content were significantly less than the other two kinds of tobacco plants.(3)In K326/solanum torvum grafting, after ridging processing induced adventitious roots of tobacco plant, leaves began to appear in the accumulation of nicotine, further demonstrates the tobacco root is the synthesis of nicotine. In adventitious roots characteristic parameters and correlation analysis found that nicotine content in tobacco and ridging induced adventitious root dry weight, root vigor were very significant positive correlation; At the same time with the increase of ridging height, adventitious root dry weight, root vigor and nicotine content were increased. Compared with the earth up 3 cm, ridging 6 cm of grafting adventitious roots of tobacco plant dry weight and root activity increased by 37. 63% and 37.63% respectively, while nicotine content corresponding increased by 32.99%.(4)The drought sensitive variety K326 as scion, drought tolerance varieties of solanum torvum as stock for grafting, in the soil relative water content of 50% of the mild drought stress and severe drought conditions of soil relative water content of 30%, K326/solanum torvum grafting showing symptoms of drought stress significantly less than K326 natural tobacco plant and K326/K326 grafting, inhibition of growth by the former process is low, root activity, chlorophyll content drop later, small amplitude, low degree of malondialdehyde content increased, SOD and CAT activity increased significantly, and under the condition of severe drought stress still maintain growth momentum.(5)Use the low temperature sensitive variety K326 as a scion, longjiang 911 as stock for grafting, low temperature resistant varieties is temperature 7? at night under low temperature stress, tobacco K326/longjiang 911 grafting showed symptoms of low temperature stress is less than K326 natural tobacco plant and K326/K326 grafting the inhibition of growth is low, small blade aspect ratio, root activity, chlorophyll content drop is small, low degree of malondialdehyde content increased, SOD and CAT activity increased significantly, and under the condition of low temperature stress is still maintain growth momentum.
