Barley yellow dwarf viruses infect many cereal plants,causing symptom development and serious damage in worldwide economy.The symptom of BYDV-GAV in wheat is dwarf and yellow.Except for that,the symptom of it on oat is reducing tillering and heading.However,now there is few study about the symptom development.For controlling BYDV-GAV disease and understanding symptom development,it is very important to study BYDV-GAV.The BYDV-GAV MP was previously considered to bind the viral RNA,increase the plasmodesmata aperture and transport viral genome.However,there are less study about MP on plant physiology.Therefore,the affect about plant physiology caused by BYDV-GAV MP was researched in this study.The main results were as follows:1.Two transcript factors,vascular plant one-zinc protein 1(AtVOZ1)and vascular plant one-zinc protein 2(AtVOZ2),were found to interact with MP by yeast two-hybrid assay.and bimolecular fluorescence complementation experiment(BiFC).According the domain of MP,three mutants MP(DH1)?MP(DH2)?MP(DH3)were constructed,the results of BiFC showed that only MP(DH2)did not interact with AtVOZ1 and AtVOZ2.It means that the N terminal 1-81aa(nuclear envelope-targeting domain)of MP were essential for the interaction;2.AtVOZ1 and AtVOZ2 were two key genes in the flowering pathway.Transgenic Arabidopsis thaliana plants that constitutively expressed MP were produced to explore whether the transgenic plant show any phenotype about flowering.Compared with wild type of Arabidopsis,Transgenic Arabidopsis thaliana plants showed more rosette leaves and more time to flowering.So MP transgenic Arabidopsis exhibited a late-flowering phenotype;3.AtVOZ1 and AtVOZ2 were genes in phyB pathway about flowering.To explore whether the interaction between MP and AtVOZ1?At VOZ2 in transgenic plant through affecting genes in phyB transduction pathway to induce late-flowering,quantitative real-time PCR(qRT-PCR)were performed.The results showed that MP transgenic plant may be through affecting the expression of FT to delay flowering.In conclusion,this study showed that BYDV-GAV MP targets at AtVOZ1 and AtVOZ2 and may through affecting the expression of FT to delay flowering,which will set foundation for the study about symptom development caused by BYDV-GAV.
