NAC family of transcription factors are numerous and widely exists in terrestrial plants, is a kind of plant specific transcription factor family, have a variety of biological functions such as involved in plant growth and development, hormone response, and many kinds of adversity stress response, etc.This paper takes Chinese Academy of Tropical Agricultural Sciences, biotechnology laboratory of coconut research institute cultivating of hainan local oil palm (Elaeis guineensis) seedlings as the research material, To study the oil palm NAC transcription factors. By bioinformatics, real time quantitative PCR analysis and Alternative splicing to study its structural characteristics and biological function. The main experimental contents are as follows:1. Multiple sequence alignment and constructing phylogenetic tree. After sequencing transcriptional income 82 oil palm NAC gene transcription the sequence search comparison genome sequence form the biotechnology laboratory of coconut research institute, Chinese Academy of Tropical Agricultural Sciences, we performed multiple sequence alignment and construct the phylogenetic tree.The results show that 82 oil palm NAC transcription factors can be divided into ?-? 6 subfamilies, each subfamily are different.The oil palm NAC gene subfamily 1 contains the most, There are 28, accounted for 34.15% of all genes; Oil palm NAC gene ? subfamily contains at least, only 6, Accounted for 7.32% of all genes.Search the 82 oil palm NAC gene protein amino acid sequence in the conservative region, make sure all oil palm of NAC genes all protein by NAM family proteins and has a specific domain NAM conservative structure, area near N end, conforms to the characteristics of highly conserved NAC transcription factor N end. The structural analysis of 82 oil palm NAC gene, chromosome location.2. We have made real time fluorescence quantitative for 32 genes that the genes to response abiotic stress by PCR amplification. Found oil palm NAC genes in response to cold, drought and salt stress the most of the total amount of gene expression in the four reaction stage showed a rising trend, a total of 21 genes, accounted for 65.63% of all genes. Of all the genes that in response to the three kinds of stress when the gene expression of overall showed a rising trend:in the cold stress, the gene that highest expression level was the highest number in 24h, a total of 10 genes; In the drought stress, the gene that highest expression level was the highest number in 48h, a total of 23 genes; In the salt stress, the gene that highest expression level was the highest number in 48h, a total of 12 genes. The experimental results show that the NAC family has a close relationship with the oil palm adversity stress, may also respond to adversity signaling molecules, induce the protein expression, resistance to stress.3.There are 20 genes have 33 different alternative splicing in 82 oil palm NAC genes. Using the 33 genes sequence design primers, with three kinds of stress processed oil palm cDNA for PCR amplification, The results showed that EgNAC06 and EgNAC45 had obvious amplified band in the cold, drought and salt three kinds of stress each processing time; EgNAC22 second alternative splicing mode and EgNAC33 second alternative splicing mode had obvious amplification bands in the cold, drought and salt three kinds of stress Part processing time; the remaining gene splicing way in the cold, drought and salt stress of all processing time period did not show significantly different amplification bands. These results show that these increase selective shear may not affect and change the oil alm NAC genes functions for response to cold, drought, salt, etc, these increase alternative splicing mode most is nonsense splicing mode.All of the above analysis and experimental results show that:oil palm NAC transcription factor may be involved in the stress response pathway when oil palm suffer from abiotic stresses such as cold, drought, salt and so on, and they can enhance the ability to resist the stress of oil palm through regulate and control the relative expression of these genes. Therefore, when the oil palm in response to abiotic stress, NAC transcription factors in regulating the oil palm adversity stress related gene expression may play a role in the reaction mechanism. This experimental research content for the oil palm NAC transcription factors in the regulation function of stress reaction and regulating mechanism provides a certain theoretical basis for further research, has a certain practical significance.
