High temperature will inhibit most plant growth and development. In order toanalysis the function of HSF family genes in rice under heat shock stress and utilizethis experimental results to obtain new rice cultivar which can tolerate heat shock stress,we use rice (Oryza. sativa L. cv. Nipponbare) to be our materials.26OsHsf genes werecloned and sequenced correctly. In order to analysis of the isoforms of OsHsfA2a(LOC_Os03g53340)generated by alternative splicing, we constructed transcriptionactivate expression vector and transcription inhibit expression vector.These26genes were cloned from the rice seedling treated with heat stress at42℃for30min and1h. It indicates that HSFs may act as an important role in heat stressresponse. Alternative splicing (AS) which creates multiple mRNA transcripts from asingle gene is known to contribute to proteome diversity and gene regulation ineukaryotes. OsHsfA2a(LOC_Os03g53340), a gene encoding heat shock transcriptionfactor (HSF) in rice, has generated six transcript isoforms (OsHsfA2a-1to OsHsfA2a-6)by AS. In this study, real-time quantitative polymerase chain reaction (RTqPCR)analysis showed that the six isoforms have different tissue-specific expression patternsunder normal growth conditions in rice. These results indicated that alternative splicingof OsHsfA2a may play an important role not only in rice growth and development butalso in regulating stress-specific cellular adaptation responses. And, stress-responsiveand phytohormone-responsive cis-elements were analysised in the promoter regions ofOsHsfA2a gene.
