The plant disease caused by pathogenic fungi could result in severe loss of agricultural production. However, both traditional methods and chemical pesticides showed great drawbacks in prevention of pathogenic fungi. Biological control draws more and more attention by its sustainable features. To collect strains with great potential of biological control, it is necessary to isolate microorganisms from different environment and build a resource library with rich species of microbia. In this study, we collected samples from different environments, including high temperature stacking, frozen soil from low temperature regions, saline-alkali soil and soil around diseased plants, and separated bacteria, fungi and actinomyces strains. We established a microbial strains repository which is able to adapt to different environments.This repository contains 175 strains,including 42 actinomycetes,28 fungus and 105 bacterials.The establishing of the repository will provide convenience for finding a rapid evaluation system.We aimed to apply this repository to screen the strain(s) with objective characteristics, i.e. strains with features of salt-resisting, heat-resistanting, cold-resisting, bio-control strains and plant pathogens.Much efforts have done to discover the bio-control microorganisms, but little is known about the mechanisms of life styles in soil and colonization style on the plants. Our lab obtained a strain from marine silt(Streptomyces albidoflavus W68), which showed antagonistic effects on several plant pathogenic fungi. We used this strain as a starting strain,and used the green fluorescent protein labeling method and aimed to trace this strain in rhizosphere soil and root,for rearch how W68 play a role in rhizophere. In this study, we built a transformant labeled by Green Fluorescent Protein based on W68 strain successfully, and made a tentative exploration by tracing the W68 in the rhizosphere soil and root of wheat. We found that this transformant could be used to research the dynamics of W68 in rhizosphere.Our works provide technical methods and theoretical basis for studying how the bio-control-like strains colonized in the roots to protect plant, and what's more,offer help for the preparation of biocontrol agent.
