Dendrobium officinale Kimura et Migo is a perennial and epiphytic herbaceous plant which belongs to Dendrobium orchid and grows on rocks of high altitudes mountains. It has been consideres as a precious Chinese herbal medicine for high medicinal value of Dendrobium officinale, which played the role of anti-aging and enhance immunity. However, due to long-term excessive excavation, Chinese Dendrobium officinale wild resources is on the verge of exhaustion, resulting Dendrobium officinale become a national second rare and endangered plants. Studies have proved that Dendrobium officinale PLBs had similar biochemical as well as physiological metabolic and morphologic development potential with original plant.Therefore, the large scale of Dendrobium officinale PLBs cultivation can replace original Dendrobium officinale, which could be an effective way to solve the the shortage of Dendrobium resources.Scientists have formed a consensus on the medicinal value f Dendrobium officinale, and the polysaccharide is the main composition of Dendrobium officinale medicinal function. A thick and short stem and a strong and viscous taste were considered as main quality of good Dendrobium officinale by Traditional Chinese Medicine experts. The content of polysaccharide are nearly the same between Dendrobium officinale PLBs and orginal wild plant, which made the cultivation of Dendrobium PLBs a possible way to offer new drug resource to develop the Dendrobium officinale polysaccharide according to limited wild Dendrobium officinale resources.This thesis has studied the following questions:the optimization of PLBs proliferation and the measurement of polysaccharide content in vitro culture of PLBs, the optimization of PLBs' industrial breeding with the improved proliferation medium, the effects of different LED light on the growth of Dendrobium officinale sprout from tissue culture, and the best proliferation medium of Dendrobium officinale sprout in land transplant. Thesis aims at establishing industrial Dendrobium officinale PLBs polysaccharide production mode and exploring the best benefits of this mode. Experiment results in this thesis will lay a foundation for improving active pharmaceutical ingredients, and provide theoretical basis for industrial and standard production of active pharmaceutical ingredients, which helps find a feasible way to creat the economic efficiency of industrial production of active pharmaceutical ingredients in Dendrobium officinale.I. The optimization of Dendrobium officinale PLBs proliferation vitro culture contidionsThe experiment results in this thesis showed that the effect of Dendrobium officinale PLBs culture was the most siginificant when added 2.0 mg/L of 6-BA, while excessive concentrations of 6-BA led to small amount of differentiation of Dendrobium officinale PLBs. In conclusion, the best recipe for Dendrobium officinale PLBs proliferation is 1/2MS (KNO3,KH2PO4:0.5and 1) with 50g/L mashed potatoes. And the growth coefficient of Dendrobium officinale PLBs came to the highest at 17.62 when the ratio of KNO3 and KH2PO4was 0.5:1 in the improved MS. The experiment results also showed that adding Methyl Jasmonate had no effect on Dendrobium officinale PLBs proliferation. Among the different proportion of organic additives, the proliferation effect was the best when added mashed potatoes, and adding the mixture of 50 g/L banana and 100 g/L coconut milk could help the differentiation of Dendrobium officinale PLBs.?. The comparison of different PLBs polysaccharide content when added different hormones and organic additivesThis thesis measured and compared the different content of PLBs polysaccharide with the addition of different proportions of hormones and organic additives conditions, and found that different combinations of hormones and organic additives have a certain influence on the content of polysaccharides PLBs. Firstly, the content of PLBs polysaccharides with organic additives was more than those without organic additives, which was due to organic additives are rich with sugar, soluble protein, fiber content, fat, carbohydrates and growth regulators. Secondly, adding proper organic additives could help to increase the accumulation of PLBs polysaccharides, while different combinations of organic additives had no significant influence on the accumulation rate of PLBs polysaccharides and the total amount of PLBs polysaccharide accumulation.?. Study on the industrial production technology of Dendrobium officinale polysaccharideThis thesis studied the industrial production technology of Dendrobium officinale polysaccharide deeply. The experiment results showed that:(1) The best combination of explants disinfection was mercuric chloride for 20 minutes and washed 4 times with sterile water,which ensured a germination rate of 89.72% and a contamination rate of 7.62%. (2) Among different culture medium of Dendrobium officinale sprout, the best culture medium for rooting was 1/2 MS +AC 0.5 g/L+ 6-BA1.5 mL+2,4-D 1.5 mg/L+sugar 30 g/L. (3) This thesis compared the influence of different LED light on the growth of Dendrobium officinale sprout with the Single factor method and found that LED warm white light contributed most to the growth of Dendrobium officinale sprout. Compared with Dendrobium officinale sprout under ordinary fluorescent white light, Dendrobium officinale sprout under warm white light had higher plant height, heavier fresh weight and bolder stem. As to the root, Dendrobium officinale sprout under ordinary fluorescent white light has longer but thinner root than Dendrobium officinale sprout under warm white light, while the latter had greener leaves than the former. Blue LED, white LED and warm white LED and fluorescent lights all contribute to promote the growth of Dendrobium officinale sprout, render the color and enhance the thickness of leaves. (4)Experiments in thesis also showed that suitable transplanting substrate was composed by pine bark, coco peat and vermiculite in ratio of3:2:2, while dry sphagnum moss is not suitable for transplanting substrate.IV. The establishment of industrial production mode of Dendrobium officinale polysaccharide and economic benefit analysis of industrial production modeAccording to previous studies, the industrial production processes of Dendrobium officinale polysaccharide are as below:firstly purchase the Dendrobium officinale PLBs, secondly proliferate Dendrobium officinale PLBs, then extracted polysaccharide directly from Dendrobium officinale PLBs and finally make the Dendrobium officinale polysaccharide into products and sell them in markets. This thesis took Dendrobium officinale PLBs as raw material and established the industrial production mode of Dendrobium officinale polysaccharide. The author took the annual production of 100kg polysaccharide as an example, calculated infrastructure, equipment, raw material purchase and financial flows of factories.Studies have shown that:it took 5.7194 million RMB for disposable investment,1.0545 million for operating extract polysaccharide directly from Dendrobium officinale PLBs to build a Dendrobium officinale polysaccharide production factory; the total income of a factory would be 1.865 million thus making 0.85 million profit every year. Therefor it would take about 7 years to cover all the cost to build a Dendrobium officinale polysaccharide production factory.
