In this paper, the zhiyuan Pleurotus eryngii and zhiyuan Ganoderma lucidum were studied. An orthogonal test of L33 that factors were cooling,cryoprotectant and thawing was designed to study technology of -80? ultra-low temperature preservation of Ganoderma lucidum and Pleurotus eryngii strains.The identification fo the effect of preservation was conducted after one year of preservation. The identification of the effect of preservation included mycelia growth index,fruiting body morPHology and yield, the nutrient content,active ingredients and ISSR molecular markers.All the identification can help to find the best combintion of conditional processing Of -80? ultra-low temperature preservation.We made some conclusions as follows:(1)The influence of mycelia growth index of -80? ultra-low temperature preservation on Ganoderma lucidum are as follows:with the 4?,-20?,-80? cooling pretreatment and 10% dimethyl sulfoxide cryopretectant,thawing in -20? 4?,room temperature was the best conditions combintion for mycelia growth index.Cryopretectant and thawing way were two main influencing factors,and cooling way followed.(2)For the mycelia growth index of Pleurotus eryngii, cooling way and cryoprotectant were two main infulencing factors,thawing way followed.with the 4?,-20?,-40?,-80? cooling pretreatment and 10% glycerinum,4?-room temperuture thawing was the best conditions combintion for mycelia growth index.(3)-80? ultra-low temperature preservation has different effects on the fruit body of Ganoderma lucidum. For the pilens dameter and the thickness of the stalk,and fruit body production, Cryopretectant and cooling way were two main influencing factors,and thawing way followed.The best conditions for the production of fruit body is A2B2C3;with the-40?,-80? cooling pretreatment and 10% glycerinum, thawing in 37? water bath.For the fruiting body morPHology and production of the Pleurotus eryngii, Cryopretectant and thawing way were two main influencing factors,and cooling way followed.The best conditions for the production of fruit body is A2B2C3,with the -40?,-80? cooling pretreatment and 10% glycerinum, thawing in 37? water bath.(3)The change of the triterpenoid and polysaccharide of Ganoderma lucidum is not obvious.There is no obvious change in the total polysaccharide,total amino acids and protein of Pleurotus eryngii.(4)-80? ultra-low temperature preservation has little effects on the the genetic stability of Ganoderma lucidum and Pleurotus eryngii. ISSR amplification experiment was carried out on the mycelium DNA.The electroPHoresis analysis show only a few specific bands. Among them,3,6 and 9 processing Of Ganoderma lucidum were affected relatively largely.2,6,9 processing Of Pleurotus eryngii were affected relatively largly. As for the proportion of total specificity strip,Ganoderma lucidum is slightly higher than Pleurotus eryngii. It shows thatultra-low temperature resistance is different for different species...
