Plant virus disease is a kind of important diseases which threaten the safe production of crops, and can cause serious damage for many crops in the world. Most plant viruses are transmitted by vector insects, which will play an important role in disease epidemics. At present, controling transmission of vector insects is one of the most popular methods to effectively control virus diseases. With the rapid development of cell biology, molecular biology, immunology and so on, the interaction mechanism between plant virus genes and vector insect genes has obtained significant advances in recent years. This will promote the study of the molecular mechanism of insect vector transmission of plant viruses and provide the new ideas for the prevention and control of plant virus diseases. Based on the yeast two-hybrid results, many proteins of S. furcifera interacted with P7-1 of Southern rice black rice dwarf virus (SRBSDV). We picked up NeuP of S. furcifera for further functional study. After injection with NeuP dsRNA, SRBSDV slO was significantly reduced in the insects and inhibition of NeuP expression also resulted in a decrease in viral transmission efficiency. The main results are as follows:1. Based on the guide of MIQE, we successfully established a real-time fluorescence quantitative PCR to detect SRBSDV in vector insect, S. furcifera.2. Based on the yeast two-hybrid results. NeuP of S. furcifera was picked up, for further study. Double stranded RNA primers was designed according to the existing sequences and then NeuP dsRNA was synthetised. After testing its specificity, we injected NeuP dsRNA to the insects of S. furcifera, injected GFP dsRNA as the control, the insects were detected by qPCR after 1 d,2d,4d,6d,8 d. The results showed that the ability of getting SRBSDV by S. furcifera injected NeuP dsRNA decreased significantly.3. In transmission assay, injected GFP dsRNA as control and NeuP dsRNA as sample group, Afer injection successfully, the injected insects were allowed to feeding on SRBSDV diseased plants for 2 days, and then trnasfer them into healthy plants one by one (single plant put a single insect). After 20 days, all rice plants were analysised results by RT-PCR. The data suggestted inhibition of NeuP expression in S. furcifera also resulted in a decrease in viral transmission efficiency (76.47%).
