In this study, the toxicity and effects of four nematicides on the insecticidal activity, locomotion and feeding behavior of Ditylenchus destructor were investigated with the method of dipping method, sand column and incubating in fluorescent cy3, the tested nematicides incluld profenofos, carbofuran, deltamethrin and abamectin.The penetrating of tow nematicides in the integumengt of Ditylenchus destructor were investigated with the method of dipping method, high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), the tested nematicide include profenofos and carbofuran. The results were summaried as follows:The toxicity and effects of four nematicides on the insecticidal activity of Ditylenchus destructor were investcided with the method of dipping method. The results showd that deltamethrin had moderate nematicidal activity against D. destructor, with LC50 value of 459.8 mg/L, while the LC50 values of profenofos, carbofuran and abamectin were 159.9 mg/L,331.9 mg/L and 257.3 mg/L, respectively.The effects of four nematicides on the locomotion of Ditylenchus destructor were investcided with sand column method.The results showd that Deltamethrin also showed better inhibitive activity on locomotion of D. destructor, with the IC50 value of 3.1 mg/L which was higher than profenofos (8.3 mg/L) and carbofuran (16.1 mg/L), but less than abamectin(0.8 mg/L).In the case of feeding, the minimal concentration of profenofos that stimulated more than 90% nematode intake cy3 was 0.6 mg/L, and 40 mg/L for carbofuran. The minimal concentration of profenofos that inhibited all of nematode intake cy3 was 360 mg/L, and 300 mg/L for carbofuran. Deltamethrin and abamectin could not stimulant the feeding of D. destructor.Abamectin can inhibited all of nematodes intake cy3 at the dose of 20 mg/L, and 200mg/L was needed in deltamethrin, both after exposure to 10 mg/L profenofos for 2 h.The residue of nematicides in the nematode of Ditylenchus destructor were investigated with the method of dipping method and high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS).The results showd that nematodes rinsed with deionized water when the dipping concentrations of profenophos, carbofuran was 200mg/L, the average value of detected residual amount were5.76,0.66mg/L; Detected residual amount were 4.38,0.96mg/L when the dipping concentrations was 400mg/L. And with methanol, deionized water washing, the dipping concentrations of profenophos, carbofuran was 200mg/L, the average value of detected residual amount were 2.76,0.022mg/L; Detected residual amount were 3.36,0.040mg/L when the dipping concentrations was 400mg/L.
