The Biological Characteristic Study Of New Strain ’Liu Yue Zao’ From Bud Mutation Of Pomelo

This study investigates and analyzes the current development of Guanxi pomelo industry and resources situation of Guanxi pomelo’s bud mutation germplasm.Then,the study concentrates on the biological characteristics and molecular marker identification of ’Liuyuezao’ which has a feature of early ripening bud mutation.The research results are following:1.After investigating the development situation of Guanxi pomelo industry,this study finds out that some development defects and obstacles exists,such as unreasonable breed constructure,relatively low cultivation and management techniques,as well as short deep processing industrial chain and aggravated regional ecological environment.This research makes a survey about Guanxi pomelo’s bud mutation germplasm and ascertains the latest bud mutation strain situation.There are precocious’Liuyuezao’,serotinous ’Zhangyi’ pomelo with tawny pericarp,’Hong Zhuang’ pomelo with special flower-pattern pericarp and scarlet ’Mini hongyou’ with small fruit.The recovery of these new bud mutation strains makes a great contribution to offering credible resource for popularizing new varieties,evaluating fruit quality,as well as adjusting plant construction and carrying out breeding work.Meanwhile,the recovery of these new bud mutation strains paves a good road for regulating Guanxi pomelo’s breed structure and polybasic development.2.Biological characteristics of the early ripening bud mutation are studied compared with Guanxi pomelo as the control.The results indicates that the biological characteristics and the phrenological phase of the early bud mutation are mainly consistent with Guaxi pomelo but the mature period is earlier 30-60 days than other main varieties.3.This study analyzes and compares the fruit of ’Liuyuezao’ with that of main varieties.The results show that the fruit integrated quality of’Liuyuezao’ is highly appreciated The weight is 1488.66±19.18g,the pericarp thickness is 183.65±3.78mm,the total acid content is 0.65±0.15%,the total content sugar is 9.15±0.16%,and the content of vitamin C is 42.92±0.96(mg/100g)is the highest in several varieties.4.Using the ISSR molecular marker technology,this research analyses 15 pomelo DNA samples,including ’Liuyuezao’,its graft generation and mother plant Guanxi pomelo,twelve available primers were selected from 100 primers.Amplification results showed that 152 bands were amplified by 12 ISSR primers,in which 123 bands were polymorphic,and the polymorphic ratio is 80.92%.The polymorphic bands has no significant difference between the ’Liuyuezao,and its generation which shows that the genetic material of grafting generation of’Liuyuezao’ does no change,and can stabilize its genetic characters.However,the amplified bands of ’Liuyuezao’ has significant difference with its mother plant Guanxi pomelo on the bands amplified by primers UBC818 and primers UBC868 which preliminarily indicates that’Liuyuezao’ is a new cultivar...
