The pumelo [Citrus grandis(L.)Osbeck cv.guanxi miyou] in Pinghe County planting area has reached 6 thousand hectares and the annual value of production is more than 0.6 billion, it is a pillar of the local agricultural economy. There are a lot of natural variation produced over 500 years of cultivation history. In this study, we used the new strain’Mini ni hong you’that is from bud mutation of’Guan xi mi you’as materials, and female parent’Guan xi mi you’as reference, by measuring the indicators of fruit quality such as fruit shape index, fruit weight, pericarp thickness, edible rate, juice yield, soluble solids, total sugar content, total acid content, VC content etc. to compare the difference of fruit quality between’Mini ni hong you’and female parent’Guan xi mi you’, and then comprehensive evaluation of the fruit quality of’Mini ni hong you’. By using I2-KI method and TTC method to measure both pollen viability, the impact by taking a different sucrose concentration, temperature and boric acid concentration of both pollen germination rate. By using ISSR Molecular Marker Technique to analyze the difference of the genetic material between’Mini ni hong you’,’Guan xi mi you’,’Red rou mi you’,’san hong mi you’ and’Hong Mian mi you’to make the genetic identification of’Mini ni hong you’. The main results of research are as follows:1. The fruit shape index of’Mini ni hong you’ was 0.84, the weight was 307.76g, the pericarp thickness was 3.60mm, the edible rate was 77.77%, the juice yield was 61.4%, the soluble solids content was 11.73%, the total content sugar was 8.03 mg/g, the total acid content was 4.52 mg/g, and the content of Vc was 45.95mg/100g. The fruit shape of’Mini ni hong you’is oblate, its peel and the sponge layer is thinner, weight of 307.76g was significantly less than the female parent of 1468.80g, edible rate was 77.77% higher than female parent of 70.10%, juice yield was 61.40% higher than female parent of 51.57%, soluble solids were 11.73% significantly higher than female parent of 10.57%. In the total sugar, total acid and content of Vc were no significant difference between ’Mini ni hong you’ and ’Guan xi mi you’.2. Through the different sucrose concentration, temperature and concentration of boric acid in ’Mini ni hong you’ studied the optimal culture conditions of pollen germination. The results indicated that the sucrose concentration was 20%, the temperature was 25℃, the concentration of boric acid was 300mg/L, that is the optimal culture conditions of pollen germination for ’Mini ni hong you’, as the same with ’Guan xi mi you’.3. Total ISSR primers 12 pairs were screened with clear, polymorphism and good reproducibility. Total 112 brands were amplified with clear, polymorphic and good reproducible, each primer could amplify 5.17 brands, of which 62 were polymorphic and the polymorphic rate was 55.36%. According to ISSR cluster analysis on the screened 12 pairs of primers, the result showed in the genetic distance of 0.2, it could separate 5 kinds of pomelo strains. It illustrates that the new strain ’Mini ni hong you’ is different from the female parent and other bud mutation strains, it can be used the development and utilization as new variety. The genetic distance of 0.3 as the cutoff point, the tested material can be divided into 3 categories, the first category is ’San hong mi you’,’Hong rou mi you’ and ’Mi ni hong you’, second is ’Guan xi mi you’, third is ’Hong mian mi you’. Use ISSR molecular markers to identify pomelo and its bud mutation strain is feasible.
