Accumulating evidence has shown microRNA play important roles in activation of embryonic genes,compaction of embryo,methylation of DNA,chromatin remodeling et al.Recent studies indicated that sperm microRNA are important for fertilization and preimplantation embryonic development and are potential vectors at the interface between genes and environment.During fertilization,a sperm not only contributes paternal genome to the zygote,but also deliver RNA and other substances into oocyte,such as mRNAs?microRNA?tRNAs and siRNAs.Compared with embryos derived from fertilization,the somatic cell nuclear transfer(SCNT)embryos show low efficiency due to the damage of micromanipulation and abnormal reprogramming.However,the role of sperm microRNA has been ignored.The aim of present study was to investigate the function of sperm microRNA in preimplantation development.Using miScript Single Cell qPCR System to analysis the expression of bta-miR-202 in sperm,oocyte,zygote and SCNT-embryo,the result shown that bta-mi R-202 was highly expressed in sperm.Intriguingly,bta-miR-202 was higher in the zygote of pronuclear stage than oocyte,2-cell embryo and the counterpart of SCNT.Previous study of our team has certificated SEPT7 was the target of bta-miR-202.Moreover,bioinformatics analyses shown that SEPT7 might interacted with HDAC6,which regulated the stability of cytoplasm skeleton protein through deacetylating ?-tubulin.The expression of SEPT7 was investigated in embryos of IVF/SCNT,the results shown that SEPT7 was lower in IVF-embryo of pronuclear stage than SCNT-embryo of pseudo-pronuclear stage.SCNT-embryo acquired with equivalent quantity of bta-miR-202 was shown lower expression of SEPT7,higher acetylization level of a-tubulin and less fragment than SCNT-embryo of control group.However,both of acquired and control group shown inferior than counterpart in term with IVF.In conclusion,the present study provide evidence bta-miR-202 regulate the first cleavage division of cloned embryo via targeting SEPT7...
