The Identification Of ERVs-derived LncRNAs In Goat Genome And The Preliminary Study On Their Effect Of Early Embryonic Develepment

Transposition is known as a common phenomenon which means genetic elements copy/cut and paste from one site to another in genome,these genetic elements was identified as Transposable Elements(TE).According to the transposition mode,they are divided into two categories: one is so-called DNA transposon,using DNA as the medium for transposable;the other is normalized as reverse transposon(retrotransposon),also known as reverse transcription element.The former is very rare in mammals,but the latter has been confirmed that they're playing important regulatory roles in the early embryonic development,placental development and many other physiological processes of humans,mice,cattle and some other mammals.ERV(Endogenous Retrovirous)as an important type of reverse transposon,their integration,expression have a significant impact of early embryonic development,the aim of this study was to find out the ERV elements in the goat genome that regulate its early embryo development and further we could investigate their affects on the early development.1.The analysis of goat genome was through the Reaptmasker and other software,selected more than 600 ERV sequence,constracted ERV database.Classify them,analyze their structures,calculate the proportions of each type of ERV,and position them on chromosomes.2.The goat oocytes were collected and subjected to in vitro fertilization(IVF).When the embryos developed to 2-cell/8-cell stage,single cell high throughput sequencing analysis was performed to establish a transcriptome database.3.Screening of ERV-derived transcription sequences that were highly expressed in the IVF 8-cell phase and lower expressed in the 2-cell phase,QRT-PCR was performed to detect false positive results.RNAi was performed to the influence of ERV-derived lncRNA/mRNA.
