In this study,sunflower seeds 2603 varieties were used as experimental materials,the orthogonal design method was used to optimize the IEF-SDS-PAGE's conditions(Two Dimensional Electrophoresis),such as the amount of sample,the isoelectric focusing time and the concentration of polyacrylamide gel,and two dimensional electrophoresis technology system,which is suitable for sunflower seeds as experimental material,was obtained.The sample size was 3.5mg,and the focusing time was 5h,and the concentration of polyacrylamide gel was 12.5%.Sunflower ovary with different pollination time was studied,and using the optimized two-dimensional electrophoresis conditions,differential protein analysis was carried out on the ovaries which had not been pollinated and 7 days after pollination and after pollination for about 28 days.The results showed that the number of differentially expressed proteins,which could be identified by ImageMaster-Dplatinum5.0software,were about 87,122 and 183,respectively.Moreover,13 differentially expressed protein spots were selected for mass spectrometry analysis.The results showed that there were 12 differentially expressed proteins in the formation process of sunflower seeds.The results showed that there were significant differences between the 3 proteins.These proteins are: Storage Protein 11 S Globulin G3,Glutelin,Enolase,Light-harvesting Chlorophyii-protein Complex Subunit ?1,26 S Protease Regulatory Subunit,Heat Shock Protein,Fructose Transferase,Enolase,ATP Synthase,Mitochondrial Glycine Decarboxylase,Concentration of Chlorophyll Protein complexes,Phosphoglycerate Mutase and S-adenosine Synthase.At last,the non-pollinated,3 days after pollination,7 days after pollination and after pollination for about 28 days in sunflower ovary as materials,using GC-MS,a sunflower Metabonomics Study method,analysis and qualitative and quantitative small molecule metabolites,find different pollination time of sunflower ovary metabolic differences.The results showed that the metabolites were mainly amino acids,organic acids,fatty acids and carbohydrates.The results of principal component analysis showed that there was a significant difference in the metabolism of ovary in different pollination time.When P<0.01,17 different metabolites,including valine,serine,xylose,were found in T test.
