Shaanbei white cashmere goats mainly distributes in Yulin and Yan'an cities in northern Shaanxi.The study that effects of diets supplemented with different levels of ensiled mulberry leaves on expression of PPAR?,ADIPOQ,AFABP of Shaanbei white cashmere goats have an important role in the application of mulberry leaves as feed for the goat to provide a theoretical basis.In this study,the expression of fat metabolism related gene m RNA and protein of Shaanbei white cashmere goat was analyzed by feeding different levels of ensiled mulberry leaves in Shaanbei white cashmere goat.The aim of this study was to find out the effects of different levels of ensiled mulberry leaves on the changes of fat metabolism of Shaanbei white cashmere goat,using the mulberry leaves as a new feed resources applied to the goat to provide the basic theoretical value,and to save the cost of feeding.In this study,40 Shaanbei white cashmere goats(wether)were randomly assigned to four groups(10 replicates,4 groups)with similar body weight(28 ± 1.05 kg)in 8-9 months.Daily ration reference to Chinese sheep breeding standards(NY/T816-2004),ensiled mulberry leaves were added levels were 0%,10%,15%,20%.The expression of PPAR?,ADIPOQ and AFABP m RNA was detected by real-time quantitative PCR in the subcutaneous fat,tail fat,perirenal fat,liver and dorsal longissimus dorsi muscle of Shaanbei white cashmere goats.The expression of the gene protein was detected by Western blot in subcutaneous fat,tail fat and perirenal adipose tissue of Shaanbei white cashmere goats.The results are as follows:1.PPAR?,ADIPOQ and AFABP mRNA were expressed in subcutaneous fat,tail fat,perirenal fat,liver and longissimus dorsi muscle of Shaanbei white cashmere goats.The expression of PPAR? mRNA in subcutaneous fat was significantly higher than that in other tissues(P <0.05).The expression of ADIPOQ mRNA in adipose tissue was significantly higher than that in liver and muscle(P <0.05).The expression of AFABP mRNA was consistent with PPAR? mRNA.2.The effects of different levels of ensiled mulberry leaves on the expression of PPAR?,ADIPOQ and AFABP mRNA in Shaanbei white cashmere goats were different.The expression of PPAR? m RNA in the subcutaneous fat and liver was significantly lower thanthat in the control group(P <0.05),while that in the perilymph and dorsal longissimus dorsi muscle is opposite.The expression level of ADIPOQ mRNA in ensiled mulberry leaves group was lower than that in control group,and the expression of ADIPOQ mRNA in tail fat and perirenal adipose tissue was higher than that in ensiled mulberry leaves groups(P <0.05),and the expression of AFABP mRNA was significantly lower than that of the control group(P<0.05).The expression of AFABP mRNA was significantly lower than that of the control group(P <0.05).The expression of AFABP mRNA was significantly lower than that of the control group(P <0.05),peripheral fat showed a trend of first ascending then descending.3.PPAR?,ADIPOQ and AFABP protein were expressed in subcutaneous fat,tail fat and perirenal adipose tissue of Shaanbei white cashmere goat.The expression of PPAR? in deep fat(perirenal fat)was significant higher than in shallow fat(P <0.05).The expression levels of ADIPOQ and AFABP in the three adipose tissues were similar(P > 0.05).4.The expression of PPAR?,ADIPOQ and AFABP protein in Shaanbei white cashmere goats were different.The expression of PPAR? protein in the subcutaneous adipose tissue of Shaanbei white cashmere goats was higher than that of the control group in general(P >0.05).The expression of PPAR? protein in the tail fat of goats was not significantly affected by supplemented with ensiled mulberry leaves in the diets(P >0.05).The expression of ADIPOQ protein in subcutaneous fat and tail fat of goats was not significantly affected by the addition of ensiled mulberry leaves(P >0.05).The expression of AFABP protein in subcutaneous adipose tissue was significantly higher than that in control group(P <0.05),and the expression of AFABP protein in subcutaneous adipose tissue was significantly higher than that in control group(P <0.05).Tail fat and peripheral fat showed a trend of first ascending then descending with the increase of the addition level.
