| MicroRNAs(miRNAs)size of 20~25 nucleotides,exists in eukaryotic organisms,is a kind of endogenous non-coding RNA with control functions.MiRNAs mature body from sequence longer primary transcription content through a series of related nuclease shear processing,assembly into RNA induce silence after complex(RNA-induced silencing complex,RISC),identify targets by means of complementary base pairing,and depending on the degree of complementary guidance silence complex target mRNA degradation or deter a translation of the target mRNA.Related studies show that gga-mir-1662,gga-mir-215 and gga-mir-1416 May be associated with the infection of chicken salmonella enteritidis,and gga-mir-1662 and TLR1 LA,gga-mir-1416 and BCL10 may be targeted.SNPS(single nucleotide polymorphism)is a genome DNA base mutation,the frequency is higher in the genome,meaningful variation can affect the function of the genome DNA components,the mRNA transcription level and protein expression,which affects the function of genes,lead to biological properties change or even cause disease.So the SNP is widely used in the study of population genetics(such as biological origin,evolution and migration,etc),and the study of disease genes,in pharmacogenomics,diagnostics,and play an important role in biomedical research.This study looked at the mechanism of the presence of salmonella infection in chicken colitis by the expression of miRNA and the expression of the target gene and the variation of mi RNA.With salmonella enteritidis infection 2 jining day age one hundred chickens,1 day,3 days after infection,7 day,14 day,21,28 and 35 days jejunum tissue samples collected,the experimental group and control group each 4,only after total RNA extraction group testing relevant mirna and the expression of target genes;7 days after infection take 200 jining only one hundred days chicken caecum contents and blood,were used to detect the content of salmonella enteritidis and extraction of genomic DNA,design the corresponding primers,the purpose of the amplification of micrornas fragments,sequencing,classification,the SNP loci associated with intestinal contents salmonella enteritidis content analysis.The results showed:1)different mirnas the timing of the control group and experimental group differentially expressed different,gga-mir-1662 and gga-mir-215 achieve differentially expressed in 7 days after the infection peak,the expression of experimental group volume,respectively 1.6 times and 1.6 times that of the control group.Gga-mir-1416 showed a marked increase in the age of 21,which was 1.5 times higher than the control group.2)miRNA and its potential target genes do not represent obvious target relationships at each point in time,and may be related to the complex regulation of the organism.On the seventh day of the infection,the increase in gga-mir-1662 was significantly increased,and TLR1 LA significantly reduced the target relationship significantly.Gga-mir-1416 and BCL10 differ significantly in the 21 st and 28 th days after infection,but do not show significant targeted relationships.3)different micrornas varied in the body,and the control group and the experimental group showed similar trends.Relative to the expression of different time points,1 day,3 days after infection,7 days,14 days belong to a high point in time,the 21 days,28 days,35 days are lower expression point in time.4)The incidence of enterocolitis with enteritis was associated with the discovery of four SNP site mutations associated with enteritis,rs18823870,rs312707374,rs316396584,rs314185118.Rs18823870 is located in the upstream,gga-mir-215 rs312707374,rs316396584,rs314185118 is located in the upstream,gga-mir-1416 further illustrate gga-mir-215 and gga – mir-1416 were significantly associated with salmonella enteritidis infection,can be used as a marker on enteritis of salmonella infection.This study identified four associated with salmonella enteritidis fungus infection of the headwaters of the micrornas SNP loci,gga-mir-215 and gga-mie-1416 were significantly associated with salmonella enteritidis infection,can be used as a marker on enteritis of salmonella infection. |
