Grape(Vitis vinifera L.)can be divided into monoecious and dioecious,while the grape staminate flower does not fruit?The V.quinquangularis Rehd is highly resistant to diseases and insect pests,resistant to dampness and high temperature.It has a certain area of artificial cultivation in Guangxi and other places in china.Because the staminate flower cannot blossom and bear fruit,the yield is affected in cultivation?he basic characters of male fruit are not clear,so the paternal selection of cross breeding is more blindness.Previous studies have shown that cytokinin treatment can transform male flowers In this paper,V.quinquangularis Rehd's bisexual flowers,which can fertilize,fertilize and bear fruit,is transformed by CPPU(cytokinin),and the natural state of V.quinquangularis Rehd's male flowers as amaterial.RNA-seq sequencing technique was used to analyze the sex differentiation,key transcription factors and metabolic pathways of V.quinquangularis Rehd.It provides an important theoretical basis for further revealing the molecular mechanism of sex differentiation in V.quinquangularis Rehd.The main achievements are as follows:1.At different times before the flowering period of the grapes,the male inflorescences were dipped in different concentrations of CPPU to compare the male sex conversion rates of different samples during the flowering season.The results showed that,the 30 days before the flowering period,50 mg/L CPPU treatments led to the conversion of the male strain of Vitis to produce the largest proportion of bisexual flowers,and the sex transformation rate was 96.26%.In addition,it was observed that the male plants of the grapevine which had been treated with CPPU solution before anthesis had delayed flowering period.This shows that,before flowering,the sex determination of the vitis grape can be affected to some extent by the application of CPPU.2.MRNA-seq analysis identified important genes and related metabolic pathways for sex determination in V.quinquangularis Rehd.The transformation of V.quinquangularis Rehd sample M and normal male CK male flowers as plant material,sample after high-throughput sequencing,the sequencing results through comparison and analysis of gene expression and metabolic pathway changes before and after sex differentiation.A total of 2468 differentially expressed genes were detected in all sex transformed body samples.By bioinformatic analysis,we identified 12 sex related transcription factors.The related metabolic pathways are Zeatin biosynthesis and plant hormone,signal transduction.Significant changes were also observed in genes controlling flowering time and plant growth cycles.These results suggest that plant hormones may be involved in the biological process of sex determination and pistil abortion in vitis.enrichment system to compare CK1vsM1,CK5vsM5,M0vsM1,M0vsM5,M0vsCK1,M0vsCK5,M1vsM5,CK1vsCK5.16 types of transcription factor family have significant differences among all CPPU treated downy grape's staminiferous flower,among which MADS-box,ARR-B,ERF,NAC,GRAS,LBD6 have no relation with the sex reversal.
