Kiwifruit is a new fruit with rich nourishment which favored by people and known as the "king of fruits".Qi Hong kiwi fruit is a variety of Chinese series which is jointly selected by Northwest A&F University,Chencang District Mango Workstation of Baoji,Meixian Horticultural Station and Qishan Kiwi Center.The Golden Fruit Kiwi is also named as "Hort16A",originated in the introduction of original resources of the kiwi fruit breeding program of Science and Industry Ministry of New Zealand in 1970 s.Nongda Yuxiang,as a large fruit type which was selected by seed breeding.As rare species,Qi Hong,Hort16 A and Nongda Yuxiang are not unique,but also has a very good prospects for the promotion and utilization value.In this study,explants in vitro culture were selected by topical buds,stem segments wit h axillary buds,leaves and petioles from those three different kiwi fruit type.In this study,technical of induced seedlings,proliferation and rooting transplanting were researched intensively.The effects of three cultivars on seedling,Planting technology,established a rapid propagation system for Kiwifruit tissue culture and provided technical support for industrial production of high quality seedlings.Test results are as follows:1.The best disinfection methods of stem segments among three varieties: the stem segments of Qi Hong and Hort16 A treated with 0.1% mercuric chloride for 5min has the best disinfection effect after it was treated with 75% alcohol for 30 s,the survival rate reached 50% and 60%,respectively.In comparison,the stalk segments of Nongda Yuxiang was treated with 0.1% mercuric chloride for 3 min could reach the best disinfection effect,and the survival rate reached 70%.2.The best medium for topical buds and stem segments among three varieties: The best medium of Qi Hong was MS + 2.0 mg / L 6-BA + 0.1 mg / L NAA,germination rate as high as 82.64%,the bud body was strong.The best medium of Hort16 A was MS + 3.0 mg / L 6-BA + 0.1 mg / L NAA,germination rate as high as 85.15%,the bud body was strong.The best medium of Nongda Yuxiang was MS + 1.0 mg / L 6-BA + 0.1 mg / L NAA,germination rate as high as 92.19%,the bud body was strong.3.The best medium for leaf and petiole callus among three varieties: The medium of MS + 1.0 mg / L ZT + 0.1 mg / L NAA for leaf and petiole of Qi Hong had the best the induction rate,which could reach 40.93%,and 45.02%,respectively.The medium of MS+2.0 mg/L ZT+0.1 mg/L NAA for leaf and petiole of Hort16 A had the best the induction rate,which could reach 41.71% and 47.43%,respectively.The medium of MS + 0.5 mg / L ZT + 0.1 mg / L NAA for leaf and petiole of Nongda Yuxiang had the best the induction rate,which could reach 45.01% and 50.31%,respectively.4.The best medium for regenerated buds of all these three varieties was MS + 4.0 mg / L 6-BA + 0.4 mg / L NAA + 0.2 mg / L GA3,the proliferation coefficient of Qi Hong,Hort16 A and Nongda Yuxiang could reach 5.75,6.23 and 4.91,respectively.The average plant height And of Qi Hong,Hort16 A and Nongda Yuxiang was 5.56 cm,5.91 cm and 5.43 cm.5.The best medium for rooting of three varieties: The best mudium of Qi Hong was 1 / 2MS + 0.6 mg / L IBA + 0.3 mg / L IAA,the rooting rate,average root numbe and average root length was 82.13%,7.14 and 6.22 cm,respectively.The best mudium of Hort16 A was 1 / 2MS + 0.9 mg / L IBA + 0.3 mg / L IAA,the rooting rate,average root numbe and average root length was 89.12%,10.33 and 6.78 cm,respectively.The best mudium of Nongda Yuxiang was 1 / 2MS + 0.9 mg / L IBA + 0.1 mg / L IAA,the rooting rate,average root numbe and average root length was 79.11%,6.72 and 6.18 cm,respectively.
