In this study,two wheat lines of Zheng Da1203 and Zheng Da1326 were selected as the materials,and irradiated with 60 Co ? irradiation,12C6+ ion beam irradiation and nitrogen ion implantation respectively.The effects of different physical mutagenic methods on the germination characteristics of wheat seeds were studied by investigating the germination characteristics and seedling growth of wheat seeds after irradiation.The effects of three irradiation methods on wheat seed were compared with each other.We explored the appropriate irradiation dose and hope that we can provide a reference for wheat mutation breeding.The main conclusion of this research as followed:1.The effect of different doses of60 Co ? radiation on the germination rate of wheat seeds was mainly due to the low germination rate of low-dose(50 Gy and 100Gy)radiation and the germination rate of control seeds(0 Gy),but when the irradiated dose was more than 100 Gy,the germination rate of the seeds irradiated by 60 Co ? was significantly lower than that of the control.When the irradiation dose exceeded the certain value,the seedling height and root length elongation were obviously inhibited.Compared with the control,the content of malondialdehyde in the wheat seedlings was significantly increased after 60 Co ? irradiation,which indicated that the radiation caused some damage to the cytoplasmic membrane of wheat seedling and produced some active oxygen and free radicals.At the same time,the contents of soluble protein and proline increased significantly,and the activity of POD,SOD and CAT increased with the increase of 60 Co ? irradiation,but it was significantly higher than that of control.2.The germination rate of the two wheat seeds was slightly higher than that of the control at the two doses of 30 Gy and 50 Gy after irradiation with different doses of12C6+ion beam.When the irradiation dose increased from 80 Gy to 100 Gy,the irradiation dose was larger,the lower the seed germination rate,and significantly lower than the control.12C6+ ion beam had a significant effect on the growth of seedlings.12C6+radiation significantly inhibited the seedling height and root length of Zhengda1203 seedlings,while the seedling height of Zhengda1326 seedlingsincreased with the increase of12C6+ion irradiation dose High to lower after the trend.The content of MDA,soluble protein and the proline were significantly increased in the leaves of wheat seedling treated with 12C6+ ion beam,which was significantly higher than that of control.The activity of protective enzyme increased first and then decreased with the increase of 12C6+ ion irradiation.3.Compared with the control,the germination rate of the two wheat materials after nitrogen ion implantation was significantly decreased,and the germination rate was the lowest when the injection dose was 4×1015 N+ / cm2.The growth of wheat seedling height and root length was stimulated at the dose of 1×101 5 N+ / cm2 and 2×1015N+/ cm2,but the seedling height and root length were inhibited when the dose was 4×101 5N+ / cm2.With the increase of nitrogen ion implantation dose,the content of MDA in wheat seedling increased,the soluble protein and free proline content of seedling increased significantly,and reached the highest value at the dose of 2 × 101 5 N+/cm2,the protective enzyme activity With the increase of nitrogen ion implantation dose increased first and then decreased.4.The dosage ranges of60 Co ?-irradiated wheat were 0 ~ 100 Gy.The dose range of 12C6+ ion beam irradiated wheat was 0 ~ 50 Gy.The dose of nitrogen ion implanted wheat was 1 x 1015N+ / cm2 and 2 x 1015N+ / cm2.
