Tick is a kind of blood-sucking ectoparasites arthropods that can carry and spread many pathogens so the human and livestock will be sick or died and it harms the health of human and the development of farming.Haemaphysalis longicornis is a member of Ixodidae,Haemaphysalis and widely distributed in the world with a large quantity so they are very dangerous.In their life cycle,Haemaphysalis longicornis feeds a lot of blood of host and the salivary gland secretes a variety of active substances that helps it suck blood.Study of salivary gland protein expression differences in the different periods to Haemaphysalis longicornis for salivary gland vampire molecular mechanism is very important,and then revealing its survival adaptation countermeasures is great significance.This work made use of the generation sequencing technology for the gene library of Haemaphysalis longicornis female tick of four stages that was unfed period,unmated feeding period,mated half blood full period and engorged period.We obtained 87825 unigene and more than 1 Kb unigene have 13222 and we got 29109 unigene that in the function annotation.And the transcriptome was used for the study of all proteins quantitative proteomics of four stages as database.We used for the iTRAQ reagents marking for the study of quantitative proteomics and the object of study was the salivary gland proteins of four stages.In the study,we got 3223 differentially expressed proteins there were 3164 were annotated and 59 were unannote.We were analysis the differential expression proteins by the bioinformatics method and found many important proteins that associated with the vampire and transport.Some proteins that we have chosen maybe have the function that affect the feeding behavior of ticks,such as kinesin,dynein,clathrin,sorting nexin,galactokinase,and adenylyl sulfate kinase,so we paid attention to the RNA interference to every protein.Finally,we found that ticks early to fall off or died because of two proteins absenced.The paper devotes to the quantitative proteomics study of different periods of salivary gland protein of Haemaphysalis longicornis and we found the kinesin and dynein that associated with the blood of female ticks and that are reproted the first time in Haemaphysalis longicornis.The experimental results have important significance to clarify the molecularmechanism of physiological changes of salivary glands and the molecular mechanism adaptive sucking in Haemaphysalis longicornis,and it is of great significance to study the control of blood sucking behavior of ticks with molecular techniques.And it provides an theoretical basis for the prevention and control of ticks.
