Establishment Of Propagation Technology For PCNA Rootstock Xiaoguo Tianshi

Xiaoguo Tianshi(Diospyros kaki Thunb.?Xiaoguo Tianshi?)is a potential excellent rootstock for the main varieties of pollination constant non-astringent(PCNA)propagation,which have shown a good compatibility with ?Taishuu?,?Matsumoto-wase? and other PCNA cultivars.However,Xiaoguo Tianshi did not have the apomixes,the effects of different pollen on their genetic stability and root system obviously.The purpose of this study is to explore the technology and methods of producing the Xiaoguo Tianshi self-rooted rootstock through the tissue culture,mound layering and hardwood cuttings.In addition,the pollinizer was investigated to obtain the effects of different pollen on Xiaoguo Tianshi,root system and other economic traits.Establishment of Xiaoguo Tianshi large-scale production technology system to provide a scientific basis.The main results are as following.1.The rooting rate was 40.5% under the condition of the optimum root medium 1/2MS(1/2N)+ sucrose 30.0 g/L+ agar 7.0 g/L+ IBA 1.0 mg/L+ PVP-40 0.6 g/L and with darkness culture for 10 days and then transferred to a medium without IBA.2.The results showed that the suitable rootstock age was around 3 years old,while the rooting was poor by using 7 years old Xiaoguo Tianshi.The girdling degree and IBA concentration presented interaction effects.When using the full girdling,the feasible concentration of IBA was 1000 mg/L;while for the half girdling,the optimum concentration of IBA was 2000 mg/L.3.Xiaoguo Tianshi hardwood cuttings could generate root on the substrate with high moisture and good air permeability.The root system performed well.In addition,the treatment of no-girdling and water for 24 hr was best for promoting Xiaoguo Tianshi hardwood cuttings to generate roots.4.“Xiaoguo Tianshi ×Male 8” showed the highest percentage of fruit setting(77.1%),average fruit weight 41.9 g,with 7 seeds per fruit,as well as the seed length and the seed width.In addition,Male 8 could help increase the root surface-area and root volume,together with the root activity of F1 seedlings.In summary,critical factor in tissue culture,mound layering and hardwood cuttings were optimized for obtaining better parameters in Xiaoguo Tianshi self-rooted rootstocks propagation.Those techniques could be adopted for further persimmon nursery industry in practice.In addition,“Male 8” exhibited the potential as best pollinizer for Xiaoguo Tianshi in sexual propagation.
