| The development of Pleurotus ostreatus(oyster mushroom)has the far-reaching impact not only on the deep processing of Agricultural Product,but also the own core cultivated breeding processes.The lectins are a large set of non-immunity-origin proteins or glyproteins which can agglutinate cells.The characterized and purifed lectins were applied in the medical field as immunity regulation,the repression of virus proliferation and the filed of antitumor.It also has a tight relation with the development of edible fungi.Here,we used the de novo approach to assembly the transcriptome of four different developmental stages of Pleurotus ostreatus strain P739 obtained by RNA-seq.Anyway,the lectin genes of P739 and genome-sequenced PC15 were analysed by bioinformatics tools based on structure and evolution.It led us to a new sight of structural and functional prediction and alternative spilicing,polyA denylation of expressed gene in specific stage by the level of transcripts.The results were listed below:1)19441165680nt bases are generated totally.For the reason that the genes of PC15 with genome sequenced show homologous less than 30% compared with those from P739.The De novo assembling approach was used.A total of 51131 Unigenes were detected,total length for unigenes is 111850405 nt,average length is 2188 nt,N50 is 3087 nt.For function annotation analysis,we got 37955,13350,25930,25621,20175,13816 unigenes which annotate to the NR,NT,Swiss-Prot,KEGG,COG,GO database,respectively,the total annotation unigenes are 38475.For protein coding region prediction analysis,the number of CDS that mapped to the protein database is 37852,the number of predicted CDS is 1536.The total number of CDS is 39388.SSR is 3581.We evaluated the quality of sequenced data,and demonstrated the specified distribution of DEGs in different developmental stage of P.ostreatus.2)On the basis of previous studies of our laboratory,we speculated the tertiary structure of protein of the lectin gene(HQ606464.1)by the methods of Homology modeling with the putative amino acid sequence.The tertiary structure of such protein is highly homologous to the lectins from higher plants and other kinds of fungal species.Then the 5 transcripts(48 transcripts before filtered for complete CDS)by RNA-Seq with start codons and stop codons annotated as lectin or lectin-like were selected to aligned to the dataset of PC15 in JGI(p<0.001).The 7 lectin genes with full length genomic DNA were discovered.Then a bioinformatics analysis based on gene&protein structure,functional motif and evolution was carried out for demonstrating the potential evolutionary relationship of these lectin genes and their potential functions,combined with the partial results such as differentiate gene expression analysis of the former section. |
