In this research,Rosa rugosa seeds as the research object,the changes of endogenous hormone contents during the development of Rosa rugosa fruits were measuring to investigate the relationship between endogenous hormones and fruit development and seed germination of Rosa rugosa.The influence of concentrated acid,seed maturity,GA3 and low temperature(4?)on seed germination of Rosa rugosa was researched under aseptic condition,through the aseptic germination of Rosa rugosa seeds.By exploring the changes of nutrients and endogenous hormones during aseptic germination process of Rosa rugosa seeds,changs of material metabolism and physiological and biochemical responses of Rosa rugosa seeds during aseptic germination process were researched.The results are as follows.ABA,IAA and GA3 existed in the Rosa rugosa fruit(pulp,seed coat and embryo)during the whole growth period.At the late stage of fruit development,the contents of ABA and IAA increased obviously,while the content of GA3 decreased.There was a germination inhibitor abscisic acid in the rose fruit,and the contents of abscisic acid(ABA)was higher in pulp and seed coat,but less in embryo,and the contents of IAA in Rosa rugosa pulp and embryo were higher in the later stage of fruit development.It indicated that the dormancy of Rosa rugosa seeds might be related to the contents of ABA,IAA and GA3 in different parts of fruit at different stages of growth and development.When 60 ds after pollinated,the seeds were mature,and with the further development of seeds,the dormancy degree of seeds gradually increased,and the time of low temperature sand storage required for breaking dormancy became longer and longer,and the germination rate was getting lower and lower.The dormancy degree of Rosa rugosa seeds is positively related to the fruit development process,which may be related to the increase of ABA and IAA contents in the fruits of Rosa rugosa and the decrease of GA3 content.During fruit development of Rosa rugosa,The ratio of ABA/GA3 and IAA/GA3 in pulp,seed coat and embryo increased gradually,The highest value reached at 120 d after pollination.And maybe that the high ABA/GA3 and IAA/GA3 ratio led to the dormancy of Rosa rugosa seeds.Aseptic germination can significantly shorten the germination time of Rosa rugosa seeds and raise its germination ratio.Before inoculation,concentrated acid treatment greatly increased the germination rate and reduced the contamination rate of the seeds,and the best treatment is concentrated sulfuric acid treat seeds 4min.The higher the degree of maturity of seeds,the lower the germination rate would be,and the best time for seed to aseptic germination is 60 d after pollination.The addition of GA3 in 1/2MS medium could promote seeds germination better,the germination time decreased and the seed germination ratio increased gradually as the treatment time at 4?lasted longer,and the best Culture condition is 1/2MS+0.15mg/LGA3,placed in 4?for 15 d after,then in the 22? light culture.Compared with the low temperature sand reservoirs in the Rosa rugosa seed during germination,significant changes occurred in the content of storage materials.Along with the prolonging of germination time,soluble protein and soluble sugar content increased gradually in the initial stage of culture,which decreased after the stage because of the utilized by embryo;the starch content decreased;amylase activity increased gradually.It showed that the storage material continued to carry out the metabolism and transformation in the culture process and provided energy for seed germination.During aseptic germination Rosa rugosa seeds,The contents of GA3 in seed showed an upward trend,whlie The contents of ABA and IAA decreased gradually,and the ratio of ABA/GA3 and IAA/GA3 decreased gradually.However,changes of hormone content of the seeds with low temperature stratification were not obvious,and the germination rate were low.It shows that aseptic germination can quickly break dormancy of Rosa rugosa seeds,and the changes of the content and ratio of hormones relate to the dormancy and germination of Rosa rugosa seeds.
