This paper took the seeds harvested in the same year as experiment material.Seeds were hidden in sands in layer,and the process of product development can be divided into the following rooting stages: before the imbibition,imbibition completely(the fresh weight of seeds was stable),the thief(seed hypocotyl broke through seed coat)and main root elongation(seed hypocotyl growth length about 30 mm),GA3 treatment(seeds were soaken in the concentration of 100 mg/L GA3 solution for 24 hours),4 ?treatment(placed the seeds in 4 ? for 14 days),epicotyl stirring(epicotyl red stirring).Determination and takes root in each stage of the sampling,three different structures: seed coat,endosperm and embryo,endogenous hormones in the seed structure,the content of GA3,IAA,ABA,ZR and total protein content in three different structures,at the same time,with film scanning,paraffin section of the seeds in different stages to take morphology and cytology observation.Test of the main results were as follows:1.The changes of endogenous hormone in seed at different rooting stages in different structures.The content of ABA and IAA in seed coat were higher than that of GA3 and ZR.ABA imbibed from the former to the epicotyl stirring stages basically stable,and the content was higher than others,as the inhibition of ABA is a kind of main germination of endogenous substances,compared with the seed coat on seed germination of mechanical obstacles to than,it may be the important reasons for seed dormancy and germination.Analysis of endosperm in the change of hormone,the rooting stage,the content of ABA content was highest,the second was IAA,GA3 and ZR contents are the lower.Embryo hormone was change in trend taking root in the last few stages whole the endosperm was consistent.The content of ABA was still the highest.Auxin IAA enhanced to promote the cell elongation,under the dual function prompted the hypocotyl elongation.In epicotyl stirring stage,the IAA has significantly increased,as IAA in germinating of epicotyl played a key role in promoting that,but at this stage the content of ABA was also having a substantial rise,it might be that the germ of the inhibitory effect of ABA.The change trend of GA3/ABA and IAA/ABA in different structures were similar with the content of GA3 and IAA overall,stated that the relative content of GA3 and IAA playing a key role in promoting taking root in different stages in different structures of for chickenin ABA Dan seed dormancy breaking and epicotyl germination.In different stages to take root,feng Dan seed imbibition before stage is far higher than that of GA3,ABA content that may be because at the beginning of the seed germination,ABA dormancy induction and maintenance of the effect,the main effect of inhibiting germination hormone;In imbibition completely,its content has not declined significantly,this may instead of to the synthesis of ABA in the process of imbibition for seed dormancy.At the same time in phoenix Dan seed root elongation stage,GA3 and IAA content increased,showed that GA3 to break dormancy and promote root play an important role,it also proved that in the process of seed germination,ABA and GA3 role has antagonism.At the same time,the auxin IAA and GA3 have synergy to promote rooting effects.2.The characteristics of total protein content at different rooting stages in different seed structures.At different stages,the content of total protein in endosperm declined gradually,it stated that energy consumption is mainly provided by endosperm in seed germination rooting process.The content of total protein in embryo,at imbibition completely and before imbibition stage,significant declined,showing that at the process of water absorption expansion,embryo itself not only consumed a lot of energy,but also absorbde the protein in the endosperm.It was a kind of transfer mechanism for providing enough energy for the germination of seeds.The content of total protein in seed coat was lower compared with other two kinds of structures,it decreased in imbibition completely phase,significant increased in white phase,it might be,in the process of water absorption expansion,seed coat was activated,seed coat came from hard dry to relatively soft jade-like stone embellish,it was also a kind of energy consumption in skin cells.3.The observation for seed morphology and cytology at different rooting stages.At white stage,embryo developed fast,the growth speed of growing point was accelerated,the development and differentiation of germ,hypocotyl,and radicle was gradually obvious,the division and elongation of radicle cells,promoted the hypocotyl elongation.As the cotyledon cells gradually spliting,the size of cotyledon gradually increasing,the accumulating of nutrients,provided the energy for the seeds to sprout.Endosperm cells,from the before the imbibition to the main root elongation,at first were long and thin flat,after swelling,cells came into the active state,providing energy for seed germination.
