Rice(Oryza sativa L.)is one of the most important food crops in the word and more than half of the population in China live on rice.Oryza longistaminata,originated in Africa,belongs to AA genome just like cultivated rice.In the long-term evolution process,it has developed a lot of good agronomic traits.Oryza longistaminata is a precious germplasm resource and a very important gene pool that can continually provides us valuable genes for the modification of cultivation rice.In this study,interspecific hybrids were obtained derived from the cross of cultivated rice Huanghuazhan and Oryza longgistaminata by the technology of embryo rescue.F2 segregation population were built by F1 mandatory selfing.And the date of population traits in Hainan Lingshui and Yangzhou are used for QTL preliminary mapping.The results are as follows:1.Thirteen interspecific hybrids derived from the cross of cultivated rice Huanghuazhan and Oryza longgistaminata were obtained with embryo rescue.The pollination success rate was 41.93%,and the mating rate was about 20.97%.F2 segregation population,including 156 individual plants,had been built by F1 mandatory selfing.2.Interspecific hybrids were transgressive inheritance in flag leaf length,flag leaf width,number of spikelets per panicle and tillers and other agronomic traits.Plant height and panicle traits prefer paternal Oryza longgistaminata.3.Based on sequence comparison between indica and japonica,we developped 593 SSR molecular markers.And there were 254 polymorphic markers between parents.The frequency of polymorphism was 42.83%.In the relatively sparse region of the chromosomes,we designed 98 Oryza longgistaminata specific Indel markers.And there were 44 polymorphic markers between parents.The frequency of polymorphism was 42.83%.A linkage map had been constructed by using 175 polymorphic markers,and it covered 12 chromosomes of rice.The total length was 2104.28 cM,and the average distance between the two markers was 12.02 cM.4.The F2 population was employed to evaluate the five major characteristics,including plant height,panicle length,flag leaf length,flag leaf width and flag leaf area.Based on the comprehensive analysis,all the traits showed transgressive segregated and exhibited continuous normal distributional,which suggested that these traits were quantitative traits and controlled by multiple genes.5.Six QTLs of plant height had been mapped in two different environments.The results showed that they located on the chromosome 1,3 and 4.The additive effect ranged from 9.42 to 21.88,and the range of individually explaining phenotypic variation was from 5.61%to 10.07%.Only a QTL was detected repeated.6.Nine QTLs of panicle length had been mapped in two different environments.The results showed that they located on the chromosome 1,3,6,8,9,10 and 11.The additive effect ranged from-2.15 to 3.30,and the range of individually explaining phenotypic variation was from 4.86%to 11.74%.6.Twenty-two QTLs of flag leaf had been mapped in two different environments.These QTLs were distributed at all the rice chromosomes except chromosomes 5,8 and 9.Seven QTLs for flag leaf length.were detected with the additive effect ranges from-5.42 to 5.08,and the range of individually explaining phenotypic variation was from 3.42%to 11.85%.Seven QTLs for flag leaf width were detected with the additive effect ranges from-0.17 to 0.12,and the range of individually explaining phenotypic variation was from 3.35%to 14.06%.Eight QTLs for flag leaf area were detected with the additive effect ranges from-7.78 to 6.54,and the range of individually explaining phenotypic variation was froml.91%to 16.12%.
