Cadmium,as the heavy metals,induced the damage of peripheral blood lymphocytes(PBLs)and kidney in humans and animals,and affected the immune response.Agaricus blazei Murill Polysaccharides(ABP)could treated the toxicity damage by cadmium induced.We studied the effect of ABP on PBLs and kidney in cadmium-induced chicken from the aspect of apoptosis,TLR4 signaling pathway and antioxidant.Seven-day-old healthy chickens were randomly divided into 4 groups,and each group contained 20 males.The cadmium-supplemented diet group was fed daily with full feed that contained 140 mg cadmium chloride(Cd Cl2)/kg and 0.2 m L saline.The ABP diet group was fed daily with full feed with 0.2 mL ABP solution(30 mg/mL)by oral gavage.The cadmium-supplemented plus ABP diet group was fed daily with full feed containing 140 mg Cd Cl2/kg and 0.2 m L ABP solution(30 mg/mL)by gavage.The control group was fed daily with full feed with 0.2 m L saline per day.At 20,40,and 60 days during the experiment,peripheral blood was collected and isolated PBLs,cell supernatant was collected after cultured for 24 h.We detected the content of cadmium in PBLs and kidney of chickens through inductively coupled plasma mass spectrometry(ICP-MS).Flow cytometry was used to detect and analysised the apoptosis rate of PBLs.The mRNA levels of Caspase-3 and Bax of PBLs and the mRNA levels of TLR4 and TLR4 signaling pathway-related factors(My D88,TRIF,NF-?B,and IRF3)of kidney in chicken by use the were analyzed by the method of q RT-PCR.The expression of TLR4 protein was detected by Western blot method.The concentrations of inflammatory cytokines(IL-1?,IL-6,and TNF-?)were measured by Sandwich-ELISA method.We detected the concentration of MDA,the activites of T-SOD and GSH-p X of kidney in chicken.The results showed that,the cadmium contents were significantly increased in PBLs and kidney,and significantly reduced after treated by ABP.The apoptosis rate of cadmium group was the highest,and was significantly reduced after treated by ABP,and the apoptosis of ABP group was significantly lower than the control group.In control group,the mRNA levels of Caspase-3 and Bax were significantly increased,Bcl-2 was significantly reduced.The Bax/Bcl-2 ratio was significantly increased.After treatment with ABP,the above changes were clearly suppressed.ABP inhibited the TLR4 m RNA level and protein expression and the m RNA levels of My D88,TRIF,NF-?B,and IRF3 in chicken PBLs and kidney,reduced the concentrations of IL-1?,IL-6,and TNF-? in Cd group chicken.The contentration of MDA in the Cd group was significantly higher than that in the control group,while the activities of T-SOD and GSH-p X were significantly lower,and the phenomenon has been significantly inhibited after treatment by ABP,the contentration of MDA significantly reduced and the activities of T-SOD and GSH-p X were significantly increased in ABP group chicken compared with the control group.These results indicates that cadmium induced apoptosis in chicken PBLs,activated the TLR4 signaling pathway of chicken PBLs and kidney,increased the expression of inflammatory cytokines,damaged the chicken kidney antioxidant function,caused PBLs and kidney damage,ABP have obvious protective effect on the above damaged by cadmium induced.In summary,our study established a model of Cd poisoning,researched the damage of Cd on chicken PBLs and kidney and the effects of ABP on cadmium toxicity.Our study provided a theoretical basis for further research on cadmium toxicity and therapeutic effect of ABP.
