This study was to investigate the relationship between TLR4/TLR7 signaling pathway and renal inflammation in avian gout induced by Nephropathogenic Infectious bronchitis virus,and to elucidate the regulation and mechanism of TLR4/TLR7 signaling pathway in renal inflammation of gout chicken induced by Kidney type IBV-virus.240 Hyline brown laying hens were randomly divided into the challenge group and the control group.On the 28 th day of the experiment,the challenge group was inoculated with Kidney type IBV-virus SX9.On the 38 th day of the experiment,the kidney tissue was collected,and the renal histopathological sections were prepared.We did some experiments,XOD,CAT,UA biochemical indicators,blood gas analysis,detection of TLR4,TLR7,MyD88,NF-κB,IL-6,IL-8,IKK,TNF-α,TRAF and other related genes in the kidney,and the results are as follows1.Compared with the control group,the renal tissue structure in the challenge group clearly showed a large number of inflammatory cell infiltration,renal structure was destroyed,fat vacuolar degeneration,and urate deposition.2.Compared with the control group,the contents of CAT and UA in the infected group increased significantly in serum and kidney,the content of XOD in serum was increased significantly,and there was no significant difference in the content of kidney.The blood gas index PH decreased significantly,and other indicators showed no significant difference.3.Compared with the control group,the gene expression of TLRs in the challenge group changed significantly: the expression of TLR4 and TLR7 mRNA increased significantly(P<0.01);the expression of TRAF-3 mRNA increased significantly(P< 0.01),the expression of TRAF-6 mRNA was significantly increased(P<0.05);the expression of NF-κB p50 mRNA was significantly decreased(P<0.01),and the expression of NF-κB p52 mRNA was significantly increased(P<0.01);IKK kinase increased only slightly;IL-6 and IL-8 mRNA increased significantly(P<0.01).MyD88 mRNA increased significantly(P<0.05),and TNF-α and IRF-7 mRNA increased significantly(P<0.01).Conclusion:In kidney-type IBV-induced avian gout,renal IBV activates TLR4 and TLR7 signaling pathways,causing changes in mRNA expression levels of related genes,activating antiviral infection mechanisms and promoting inflammation of the body,causing renal lesions.
