The purpose of the paper is to explore the mechanism of salt tolerance in forage leguminous species and to determine the suitable.concentration of salt-tolerant mutant screening from the cell level.The embryogenic callus of Medicago varia Martin.cv.CaoYuanNo.3 and a wild specie(Lotus corniculatus L.)were used as the materials,which were induced by MS callus induction medium,the tests of two species leguminous forage callus have been analyzed by the from relative growth rates,physiological and biochemical indexes(malondialdehyde content,soluble sugar content,soluble protein content,proline content and three kinds of antioxidant enzymes)under different NaCl content(0,0.2%,0.6%,1.0%,1.5%)and different stress time(5d,10d,20d,40d).The salt tolerance of callus was evaluated by membership function method.The results were:(1)Under NaCl stress,the growth and induction of embryogenic callus of Medicago varia Martin.cv.CaoYuanNo.3 and Lotus corniculatus L.wild specie were inhibited,while in the short time(5d,10d),the growth of two forage leguminous species callus was promoted with NaCl concentration of 0.2%.(2)The effects of NaCl stress on the growth,osmotic adjustment and antioxidant ability of Medicago varia Martin.cv.CaoYuanNo.3 and Lotus corniculatus L.were synthetically analyzed,the results showed that 1.0%NaCl concentration should be selected as the suitable concentration for Medicago varia Martin.cv.CaoYuanNo.3 asexual screening mutants,while the suitable concentration for Lotus corniculatus L.should be selected between 1.0%and 1.5%.(3)The salt tolerance of Medicago varia Martin.cv.CaoYuanNo.3 callus was increased when the NaCl stress concentration>0.2%.while the salt tolerance of Lotus corniculat L.could be improved when the NaCl stress concentration was>0.6%.
