Plasma membrane intrinsic proteins belongto a family of aquaporin.They have the effect of controlling the transport of intracellular water molecules,which can on the infection of plant-pathogens.Through the effect on the infection of plant-pathogens,PIPs can participate in plantimmune responses.The family of rice PIP proteins includes 11 members,which are OsPIP1;1 to OsPIP1;3 and OsPIP2;1 to OsPIP2;8.For the purpose of understanding the relevant mechanisms between rice and defend to the Bacterial leaf blight,this essay is going to study the relationships among 11 types of rice PIPs genes expression and the infect of Bacterial leaf blight,while preliminarily discuss the function of hormone signal at the same time.1.The relationship between PIPs gene expression and rice bacterial blight infectionChoose to the susceptible rice varieties Nipponbare and relatively disease-resistant rice varieties IRBB13 to test.The results show that:the expression levels of OsPIP2;2、OsPIP2;5、OsPIP2;7 and OsPIP2;8 were significantly reduced by 24%-27%after pathogen infection in Nipponbare.On the contrary,OsPIP2;3 expression levels significantly increased by 28%.The remaining gene expression unaffected.In IRBB13,expression levels of OsPIP2;7 is increased by 20%.On the contrary,expression levels of OsPIP2;2 is decreased by 26%.The remaining genes,expression are unaffected.Therefore,OsPIP2;2,OsPIP2;5,OsPIP2;7 and OsPIP2;8 closely related to Bacterial Blight susceptibility,OsPIP2;3 have a certain relationship with the disease resistance.2.Effects ethylene signal for PIPs genes expression and bacterial blight infectionAnalysis of ethylene signal effect to the rice PIPs gene expression and bacterial blight.The results show that compared with the no processing,ethylene precursor ACC pretreat to rice,which can reduced the genes expression level of OsPIP2;1、OsPIP2;4.OsPIP2;7 in Nipponbare.It can lead to reduce the genes expression level of OsPIP2;7 in IRBB13.When using the ACC with 1-MCP treatment,can reduce the ethylene signal OsPIP2;1,OsPIP2;4 and OsPIP2;7 in Nipponbare inhibition,making three genes clear expression levels increased in Nipponbare.It is possible to reduce the ethylene signal OsPIP2;7 inhibition on IRBB13,making OsPIP2;7 increase the expression level of genes.Ethylene signal transduction inhibitor AgNO3 and 1-MCP have similar effect.Bacterial leaf blight symptoms and measured the amount of bacteria breeding in rice leaves showed.ACC treatment regardless of Nipponbare or IRBB13 can increase lesion length.It has a similar effect on the amount of bacteria breeding in the blade.In contrast,in the ACC with 1-MCP or the amount of bacteria breeding AgNO3 mixing process in the leaves increased significantly,lesion length was significantly longer.Thus,ethylene signals may affect the expression level of OsPIP2;7.Thus effect the bacteria breeding in rice leaf tissue,leading to bacterial blight lesion length,thereby increasing bacterial blight symptoms OsPIP2;1 and OsPIP2;7.3.Impact of salicylic acid signal PIPs genes expression and bacterial blight infectionThe effect of salicylic acid signal rice OsPIPs genes expression and bacterial blight infection,results showed that compared with the control,salicylic acid treatment increased the expression level of OsPIP1;1,OsPIP1;2,OsPIP2;1,OsPIP2;2,OsPIP2;3,OsPIP2;4,OsPIP2;5,OsPIP2;7,OsPIP2;8 in Nipponbare.Meanwhile,the salicylic acid treatment significantly reduced OsPIPl;1,OsPIP1;2,OsPIP2;2,OsPIP2;3,OsPIP2;4,OsPIP2;5,OsPIP2;7,OsPIP2;8 expression in the IRBB13.Visible,except OsPIP1;3 and OsPIP2;6 except salicylic acid are subject to signal suppression.Salicylic Acid can reduce Nipponbare and IRBB13 bacterial blight symptoms,the amount of bacteria breeding in the leaf tissue is also correspondingly reduced.Presumably,salicylic acid signal by inducing the expression of many OsPIPs inhibit bacterial blight infection,enhance disease resistance in rice.4.Effect of jasmonic acid signal PIPs gene expression and bacterial blight infectionThe effect of jasmonic acid signal can effect the rice OsPIPs genes expression and bacterial blight.The results showed that,compared with the control,methyl jasmonate may Nipponbare except OsPIP2;6 other than the 10 OsPIPs genes are inhibited,which OsPIP1;1,OsPIP1;2,OsPIP2;1,OsPIP2;4 and OsPIP2;7 expression in Nipponbare decreased more obvious,and methyl jasmonate and NDGA common treatment,can reduce the NDGA on OsPIP1;1 OsPIP1;2,OsPIP2;1,OsPIP2;4,and OsPIP2;7 inhibitory effect of expression,such that expression of the rise.In IRBB13,after ACC processing OsPIPs gene inhibition is not obvious.The effect of NDGA on MeJA inhibited,NDGA treatment significantly reduced leaf blight symptoms Nipponbare and IRBB13 reduce the bacteria content in the leaf tissue.These results indicate that the JA signaling by affecting the expression of OsPIPs genes.They can infect rice bacterial leaf blight fungus resistance.According to the findings,more OsPIPs genes expression induced by pathogen infection.A plant hormone ethylene,salicylic acid,jasmonic acid signal OsPIPs can impact on a variety of gene expression.Thereby in fect rice bacterial leaf blight fungus resistance.In 11 kinds OsPIPs genes,OsPIP2;7 in susceptible and resistant rice varieties than the expression on Infection by bacterial blight,as well as by three hormones signals conditioning,defense responses in rice Bacterial Blight species may play an important role.
