Background: Plants infected by pathogenic bacteria can express a number of proteins,the proteins is named Pathogenesis-related (PR). At present, in nearly all reports have PRgenes in plants, which prompted that the PR genes are important. There is a large numberof experimental evidence that PR proteins play an important role in the disease resistanceresponse. But PR1frequently used as marker genes for systemic acquired resistance inmany plant species. Rice is one of the principal food,and to monocotyledonous species itis also a model organism. While the bacterial leaf blight pathogen caused by Xanthomonascan seriously affect the output of rice, precipitously. Now the cultivation of resistantvariety is the most effective way to control the disease. Xa21is a broad-spectrumresistance gene. The Xa21mediates immune response of rice by identifying the non-toxicdeterminer of pathogenic bacteria. Researching the expression features of protein in theinteraction between rice and Xoo is important to identify the Xa21disease resistance andparse the pathogen resistance mechanism. Studies have proved that PR1protein familymembers involved in the pathway of rice resistance to bacterial leaf blight.Aim: We prepared antibodies against PR1proteins, to explore expression changes ofPR1family protein in normal growth rice leaves and interactions between rice andbacterial leaf blight, to compare the differential expression of PR1family in differenttreatments leaf, then lay the foundation of continue to research the protein function.Material and methods: Total proteins were isolated from both susceptible andresistance rice leaves after infection with Xoo, and at the same time distilled water as acontrol.Then we design the polyclonal antibodies and check the expression changes ofPR1proteins at different treatments and different times by Western blotting.Results: In normal growth rice leaves, PR1#012、PR1#021、PR1#022、PR1#051、PR1#071and PR1#072express low in seedling stage and upregulate with the development;generally the hightset level is vigorous growth period, and fell slightly in mature. PR1#052and PR1#073express relatively high at seedling stage, and decline in mature period, whilewe didn’t detect the expression of PR1#121.In the Xa21-mediated resistance response to Xoo, we found that PR1#012、PR1#051、PR1#052、PR1#072、PR1#073and PR1#121present up-regulated or induced, PR1#071appear down-regulate. The abundancechanges in the resistance and susceptible is significantly greater than mock.Conclusion: This study revealed the PR1family participation the pathway ofXa21-mediated resistance response to Xoo, laid a solid foundation for the further researchthe function of PRs.
