Jinjiang cattle is the local characteristics of Jiangxi cattle,after a long period of breeding,with high temperature and humidity,resistance to coarse feeding,strong resistance and other advantages.Because the meat is delicious,nutritious,taste good,Gao'an Jinjiang beef renowned Fujian,Guangdong and Jiangxi,the market prospect is broad.In genetic ancestry,feed utilization,heat stress and feed additive as only some research reports,but is associated with high quality beef fat metabolism characteristics are not reported.Therefore,this article takes jinjiang cattle as test object,aimed at the different stages of weight jinjiang cattle,body size and blood biochemical indices and relative expression of semitendinosus muscle fat metabolism genes,the results will be for the growth and development of jinjiang cattle and scientific supplementary feeding fattening to provide the reference data.The first part:This experiment was designed to investigate the characteristics of different weight of Jinjiang cattle body size and backfat development.According to the farm situation,choose in 160 kg to 340 kg weight range of jinjiang scalpers fifteen head,divided into five groups,each group of three.For three consecutive days,every morning 8:00,afternoon 3:00,the determination of environmental temperature and humidity and calculate the temperature and humidity index;At the same time,the determination of jinjiang cattle,rectal temperature,respiratory frequency,body size and backfat.The results showed that:1)In the cowshed for 60 under the condition of average temperature humidity index,rectal temperature and respiration rate at different weight of Jinjiang cattle had no obvious change,basic consistent.2)With the increase of body weight,body size and backfat also will increase.Conclusion:Backfat,body size and weight of Jinjiang cattle were extremely significant positive correlation.The second part:This test on blood indexes of Jinjiang cattle in different weight stages were determined and analyzed.Groups as former,through the jugular vein blood collection,analysis and determination of related blood indicators.The results showed that:1)Blood urea nitrogen and creatinine concentrations were not significantly different between the groups;But the blood cholesterol,glucose,triglyceride,high/low density lipoprotein concentrations among the groups were significantly different(P<0.05).2)Serum leptin,adiponectin,insulin,growth hormone,estrogen,TNF and NADPH were significantly different between groups(P<0.05);Blood Acetyl-COA carboxylase and interleukin 6 difference between groups were not significant.3)Jinjiang cattle body weight and triglyceride,low density lipoprotein cholesterol levels were extremely significant positive correlation(P<0.01);Body weight and creatinine,NADPH content was significantly positively correlated(P<0.05);Body weight was negatively correlated with total cholesterol,insulin and leptin levels(P<0.05).Conclusion:1)There is a certain correlation between the blood indexes of Jinjiang cattle.Among them,the leptin concentrations in the blood and body weight was significantly negative correlation;Triglyceride concentrations in the blood and negatively correlated with total cholesterol concentration.2)When the weight at about 300 kg,Jinjiang cattle blood indicators have reached a higher level,suggesting that should pay attention to the stage of nutritional supplements and feeding and management in the future course of fattening.The third part:Grouping test ibid,with the help of the veterinarian,select Jinjiang cattle hind legs semitendinosus muscle tissue in vivo sampling.Using fluorescence quantitative PCR method,the content of fat metabolism related genes in the muscle tissue of Jinjiang cattle was detected,including the key genes of fat deposition:FABP4,C/EBP,PPAR? and SREBP1;Fat decomposition related genes:LPL,PPARa and UCP.In order to explore the change of fat metabolism related genes and the regulation of fat metabolism in Jinjiang cattle at different stages of weight.The results showed that:1)The FABP4 gene expression of the key gene of Jinjiang cattle was higher in 162 kg,and the expression of C/EBP and SREBP1 was significantly increased at 318 kg stage.2)Determination of fat decomposition related genes in Jinjiang cattle.It was showed that the expression of LPL and PPARa gene was significantly down regulated during the weight stage of 262 kg and 342 kg,while the expression of UCP gene was significantly increased in the weight stage 208 kg and 318 kg.Conclusion:The expression of key genes of fat deposition in muscle tissue of Jinjiang cattle at different weight stages was in a dynamic process,the weight of which is the peak of fat deposition in 300 kg,which can provide reference for the production of high quality beef.
