The fruits of Ziziphus jujube Mill cv.Lingwuchangzao in different developmental periods from different areas in Ningxia(Lingwu,Yinchuan,Zhongning,Qingtongxia)were used as the experimental material.The water extraction and alcohol precipitation method were adopted to obtain polysaccharide,which was subsequently purified by DEAE-52 cellulose column and HW-55S gel column.Finally gas chromatography-mass spectrography(GC-MS)was used to analyze the monosaccharide composition from purifed polysaccharides.The results showed that:1?With the growth and development of the fruit,the yield of crude polysaccharides from different areas showed a rising trend,and the crude polysaccharides at the maturation period from Lingwu had the highest yield,1.795%;By phenol-sulfuric acid method to detect contents of crude polysaccharides,the content was on the rise in the developmental process,the content was highest in polysaccharides at the maturation period from Lingwu,reached 59.76%.2?After purifed by DEAE-cellulose52,all polysaccharides were fractioned respectively to one neutral fraction,Ju-0,and three acid fractions,Ju-1,Ju-2,Ju-3.And Ju-2 had the highest contents.This indicated that the main components of polysaccharides in Ziziphus Jujube Mill cv.Lingwuchangzao was acid fractions.The fractions were purified by HW-55S column,and we got white refined polysaccharides.With the growth and development of the fruit,the content of refined polysaccharides from different areas showed a rising trend,which were different among different areas.In the early bulking period,the rapid enlargement period and the maturation period,refined polysaccharides from Lingwu area had highest content,while Zhongning area had highest content in the coloring period.3?The results from GC-MS showed that Ziziphus jujube Mill cv.Lingwuchangzao were composed of arabinose,rhamnose,ribose,fucose,xylose,mannose,galactose and glucose,glucuronic acid,galacturonic acid,but composition and content are different among different areas.Arabinose,ribose,galactose,glucuronic acid and galacturonic acid were the main components,and the content of xylose was the lowest.Both Ju-0 and Ju-1 were mainly composed by arabinose,ribose,while Ju-2 and Ju-3 existed obvious differences among four ares.According to the comparision of the total contents of monosaccharides,the content of glucuronic acid and galacturonic acid decreased in polysaccharides from Lingwu in the developmental process,while arabinose and ribose gradually increased;The content of arabinose was highest in the early bulking period,but the content of glucuronic acid and galacturonic acid gradually increased to the maturation period,and reach the highest.So the dynamic change of the content arabinose,ribose,glucuronic acid and galacturonic acid might be the a factor which led to the difference in the quality of Ziziphus jujube Mill cv.Lingwuchangzao from different areas in Ningxia.
